Just got back from ESMO Geneva. And wow. Immuno-oncology research is truly inspiring. Here are 3 things that stood out: 1. T Cell Engineering & Functionality Engineering T cells to overcome immunosuppression and maintain functionality in the tumor microenvironment (TME) is critical for therapies like CAR-T and TCR-T. - Advancements in armored CAR-T cells (e.g., IL-12-secreting CAR-T) and CRISPR-based editing to enhance resistance to checkpoint blockade or metabolic suppression. - Efforts to improve trafficking and infiltration into solid tumors, which remain a major hurdle. - T cell exhaustion, such as modulating transcription factors like TOX or using checkpoint inhibitors in combination. 2. Vaccine-Based Immunotherapies - Oncolytic vaccines with checkpoint inhibitors or cytokine therapies (e.g., IL-2, IL-15) could overcome "second resistance." - anti-CD25 therapies: These target Tregs (regulatory T cells), which often suppress immune responses in the TME. Exciting progress into balancing immune activation without unleashing autoimmunity. - Use of mRNA vaccines in oncology (beyond COVID-19) and how they are influencing the immuno-oncology vaccine space. 3. Neoantigen Identification & Immune Profiling - Exome sequencing is a gold mine for neoantigen discovery - computational bio making big steps with AI to find immunogenic targets. - Circulating tumor DNA (ctDNA) can be used to detect those neoantigens (although detection in blood isn't possible yet) - Several exciting neoantigen-based vaccines and TCR-T cell therapies are under development Overall, my 3 key areas to keep following: - Precision molecular tools shaping next-generation therapies. - Engineered immune cells overcoming tumor resistance. - Innovative vaccines transforming cancer care. 👥 Please join us in welcoming Carolina Almeida and Zhentao Liu to our engineering and product team, we are super excited to have them with us, as we head into 2025! 🔒 Zhentao holds a Master’s in Cyber Security from EPFL and ETH Zürich. Before joining Rivia as a Software Engineer, Zhentao gained valuable experience through internships at Logitech, ELCA Group, and Tune Insight, working on projects in full-stack development, AI , and security. Zhentao is a Chinese Go player who is actively competing in Go tournaments across Europe. 🔧 Carolina holds a Master's degree in Biomedical Engineering from Universidade de Aveiro Portugal and brings valuable experience in both tech startups and data science. She has developed innovative wearable technologies and led a small-scale clinical trial, earning academic recognition for her work. Outside of work, she's a music lover and avid vinyl collector, with a deep appreciation for the artistry and history behind the records she collects. 🌟 Last week the team gathered at the heights of Zürich to plan for 2025. Uetliberg was the perfect outlook as we worked on how we want to grow in the coming year ⛰️ During our team offsite, we focused on collective growth and acceleration. What does this mean: 1️⃣ We worked as pairs and groups on perfecting our product pitch, USP, and use cases for different client personas (not only pitching to scientists and lab experts but also our children and grandparents) - this was great fun and imaginative! 2️⃣ We worked in pairs and as a team to understand our individual and team soft and hard competencies. Using a range of competencies, we learned more about ourselves and each other, and how we can work together / communicate effectively, as the team grows. 3️⃣ Together we aimed to understand how we evolved as a company. Growing from 3 to 12 employees in a few months brought cultural changes. As a team, we defined our Guiding Principles, which allows us to better understand our cultural environment, how we collaborate, and how / who we want to bring into the tea in 2025. 👉 We had many individual and team outcomes and learnings and the biggest outcome from the offsite was one of a combined mission, we're all in this, we're invested and in it to win it - let's go team 🤩 #teamgrowth #teamoffsite #letsgo
