The development of targeted therapies for the treatment of diseases like cancer requires a deep understanding of the underlying pathogenic mechanisms. The dTAG approach highlighted in this AppNote offers a useful tool to study the involvement of proteins in a disease. It utilizes CRISPR/Cas9 to artificially introduce a dTAG binding motif into theprotein of interest. Adding a dTAG degrader molecule to this approach results in the ubiquitination and subsequent degradation of the protein. This approach allows to screen for the involvement of proteins in disease in high throughput on microplate readers.
