For as long as we can remember, our planet has generously given us everything we have needed and everything we have wanted. Yet our resources are limited. Our next steps must be chosen carefully. It is our ambition to do our best and to provide long-term as well as more environmentally friendly solutions by utilizing processes from nature. We are working on projects in which new products are created from waste materials and design new biocatalytic technologies for a more sustainable future. Through real-time analytics, detailed process control is enabled, resulting in a more responsible management of our resources and minimization of waste streams. We are digitalizing these processes to better understand them and to establish smart processes. To invent new and innovative methods, teamwork and interdisciplinary collaboration are of utmost importance. We are globally networked. Additionally, we cherish close cooperation with partners from industry, as this allows our research results to be implemented on a large scale. At the same time, this enables our students and employees to get in contact with industrial research and development at an early stage in their career. Thanks to our interdisciplinary and intercultural collaborative environment, we have even more opportunities to develop new ideas and to put our visions into practice. We do not just want to help our students become better scientists, but also give them the freedom they need to develop as individuals. To empower the future generation with new, sustainable solutions, we invite children and youths to come and experience the world of science and technology at our institute. As part of our "Kinderforscher" program, we develop and offer experiment kits to elementary and secondary schools. Happy to share our latest research article exploring the biocatalytic synthesis of sacubitril valsartan sodium hydrate, a key active pharmaceutical ingredient! This paper is the result of a collaboration between the Institute of Technical Biocatalysis - TUHH (Hamburg University of Technology) and Novartis and is authored by Zeynep Perçin, Florian Kleinbeck, Paul Bubenheim, Thomas Ruch and Andreas Liese. Here are the highlights: 🔬 Equilibrium: The reaction equilibrium strongly favors the desired product improving process efficiency. ⚙️ Kinetic Models: The reaction was modeled with both ping-pong two-substrate kinetics and Michaelis–Menten double substrate kinetics, identifying the latter as the best fit. ⚡ Inhibition Challenges: Product inhibition emerged as a key bottleneck, more impactful than substrate inhibition. 🌟 Reactor Innovation: Transitioning to a plug flow reactor proved to be a key aspect in decreasing inhibition and increasing performance. This study highlights the importance of biocatalysis to provide sustainable, escalable solutions for pharmaceutical synthesis. By understanding the detailed kinetics, we pave the way for more efficient manufacturing of pharmaceutical compounds. We are looking forward to actively participating in this Deutsche Forschungsgemeinschaft (DFG) - German Research Foundation funded RTG (research training school) on Climate Informed Engineering, excellently fitting with the United Nations University Hub at the Technische Universität Hamburg
We have received approximately 7 million euros in research funding from DFG! I am pleased to announce that our RTG proposal on Climate-Informed Engineering has been successfully funded by the DFG! We have secured approximately 7 million euros for this project. We introduced the concept of Climate-Informed Engineering first in an invited article, which made it to the cover of the ACS Engineering AU in 2023 (the photo below). This paper formed the foundation of our proposal. We plan to recruit 20 PhD candidates in this project along with a postdoc researcher. This project holds significant importance for us at Hamburg University of Technology, as it aligns with the research direction of TUHH and the focus of the United Nations University Hub at TUHH on Engineering to Face Climate Change. As John F. Kennedy once said, “Victory has a thousand fathers, but defeat is an orphan”. So, I cannot announce this great news without acknowledging the incredible individuals who made this possible. First and foremost, I owe immense gratitude to my lifelong friend, companion, and wife, Dr. Salome Michelle Sophie Shokri-Kuehni. Living with someone who works around the clock is not easy task, yet she has been nothing short of amazing. Salome has shouldered countless responsibilities, allowing me to pursue ambitious goals with determination. Her patience, strength and wisdom have provided a solid foundation upon which I could aim for the highest goal I could possibly ever imagine. Special thanks also go to the PIs of the project including Peter Fröhle, Stefan Heinrich, Cathy Hohenegger, Patrick Huber, Andreas Liese, Thomas Rung, Marcus Rutner, Michael Schlüter, Kay Smarsly, irina smirnova, and Bjorn Stevens. 🚀Excited to share the latest paper of our PhD candidate Kristin Hölting!🎉 Kristin is doing her PhD at the Institute of Technical Biocatalysis - TUHH (Hamburg University of Technology) and GALAB Laboratories GmbH and has developed a modelling approach for the continuous synthesis of N-Acetylneuraminic Acid (Neu5Ac) using packed bed reactors. 🔬 Key highlights: ✔️ Immobilized enzymes (N-acetylglucosamine 2-epimerase & N-acetylneuraminic acid lyase) demonstrated high stability, with half-lives of up to 58 days. ✔️ Kinetic studies confirmed Michaelis-Menten behavior, and the developed Python-based model achieved prediction accuracies of 94% under varying conditions. ✔️This model will be used to optimize reactor dimensions, flow rates, and substrate concentrations for cost-effective and efficient biocatalytic production. 🎉 New Publication Alert! 🧪 Excited to share that our latest paper has just been published! Luca Schmidt is a PhD candidate in the Process Analytical Technologies Group led by Victoria Bueschler at the Institute of Technical Biocatalysis - TUHH (Hamburg University of Technology). In this paper, Luca has investigated the application of a commercial low-field benchtop NMR for real-time monitoring of enzymatically catalyzed reactions. Specifically, the paper is focused on the diastereoselective aldol reaction between glycine and benzaldehyde, catalyzed by threonine aldolase. Key Findings: ✔ The complemental hard modeling (CHM) approach successfully differentiated diastereomers, even with signal overlap challenges. ✔ For the first time, thermodynamic equilibrium was observed in the aldol reaction using in-line methods at 30% benzaldehyde conversion after just 2 hours. ✔ The results show a diastereomeric excess of 37:63 (erythro/threo) via in-line NMR, closely matching off-line GC and HPLC analysis. This work highlights the potential of benchtop NMR systems to revolutionize in-line monitoring in enzymatic reactions, offering valuable insights into reaction selectivity, mechanisms, and equilibrium dynamics.
