Conventional intestinal screening models do not reflect the complex physiology of the human intestinal tract, preventing many drug candidates from reaching clinical approval. 🎯 To better predict oral bioavailability and intestinal drug metabolism, the UNLOOC Chips JU Project aims to develop an organ-on-chip system combining liver, gut, and gut microbiota in a multiwell plate format, with non-invasive sensors to monitor their activity in real-time. Regarding the biological component of this multi-OOC: ✅ Tissue-derived intestinal stem cells will be obtained by TNO, cultured as organoids, and an epithelial barrier model in an OOC device. ✅ Institute for Bioengineering of Catalonia (IBEC) will produce 3D microlivers in a biodegradable gel using either immortalized human hepatocytes (HepaRG) and stellate cells (LX2), or primary human hepatocytes. ✅ BeCytes' contribution will be essential to providing human hepatocytes from 20 donors for developing these minilivers. ✅ Human liver spheroids produced by InSphero will be applied for the final pharmaceutical validation.
