Celtarys Research develops and commercializes new chemical tools to spread the use of fluorescence-based methods in drug discovery. Our chemical conjugation technology allows us to develop in a competitive manner and short time (<3 months) customized fluorescent ligands with optimal pharmacological and photophysical properties for any druggable target. We have initially applied our technology to develop versatile fluorescent GPCR ligands which we have externally validated in several HTS assays such as FRET, Fluorescence Polarization, High Content Screening or Flow Cytometry, avoiding the use of radioactivity. Products: We have a catalogue of 20 fluorescent ligands for different families of GPCRs, including adenosine, dopamine, serotonin, cannabinoid and muscarinic receptors. Compared to other available fluorescence-based technologies, our ligands are: -More flexible and efficient -With high affinity and selectivity for the target -With a wide variety of fluorochromes -Compatible with various assays: FRET, polarized fluorescence, High Content Screening, flow cytometry... -Rational design portfolio covering entire GPCR families - With low cost and scalable production Services: We also offer custom synthesis of specific fluorescent ligands for any target with therapeutic potential.   • One product, many faces... the story behind Luxendin. New fluorescence tools for GLP1R characterization. Just like every person, each product has a story behind it. At Celtarys Research, we believe it's important to take a moment to reflect on the effort and passion that go into creating our products. The glucagon-like peptide-1 receptor (#GLP1R), a class B G protein-coupled receptor (#GPCR), plays a crucial role in regulating metabolism. Nowadays a key part of diabetes type II treatment, it has long been a challenging receptor to characterize, with numerous research groups investing significant time and resources into its study. To remark on its importance, in the last five years alone, there have been over 4,000 publications and more than 200 clinical studies dedicated to understanding this receptor. Historically, visualizing #GLP1R has been limited to methods like genetic manipulation, antibody-based assays, or the use of probes that stimulate receptor activation. Despite these challenges, two groups—led by David J. Hodson and Johannes Johannes Broichhagen, in collaboration with numerous other researchers—set out to develop new #fluorescent tools to better characterize this intriguing receptor and support #pharmacological research. Their first creation of this joint effort was LUXendin645 (#CELT-112), a far-red fluorescent GLP1R antagonistic peptide label, capable of specifically labelling GLP1R on membranes in both live and fixed tissues. These tools have significantly advanced our ability to characterize GLP1R expression in brain and hESC-derived β-like cells, revealing that the GLP1R family exhibits higher-order organization, including the formation of conserved membrane nanodomains. Despite their great achievements, the detection and visualization of GLP1R has remained challenging, partly due to the limited availability of validated reagents. In light of this, the two leading groups recognized the importance of developing a broader range of fluorescent probes with diverse wavelengths. They decided to focus their efforts on generating probes covering the green and near-infrared color ranges, successfully synthesizing and testing LUXendin492, LUXendin551 (CELT-111), LUXendin645 (CELT-112), and LUXendin762 (CELT-113) used to specifically label GLP1R in cells and pancreatic islets. Thanks to the incredible efforts of these researchers, Celtarys can now offer a wide range of fluorescent ligands targeting GLP1 (CELT-111, 112, & 113) (https://lnkd.in/ddkuJbrb) with various wavelengths. The packaging format of our product (3 nanomoles) is designed to provide the correct dosage for preclinical rodent studies through injection.

  • Luxendin #GLP1 has now become more accessible to all researchers worldwide. This is possible thanks to the commercialization agreement between Celtarys Research, leader in the development of new fluorescent tools for drug discovery, and Max-Planck-Innovation GmbH, Leibniz-Forschungsinstitut für Molekulare Pharmakologie (FMP) and University of Birmingham. CELT-111, 112 and 113 (https://lnkd.in/ddkuJbrb), high-affinity #GLP1R fluorescent antagonists, were developed through the joint collaboration between the laboratory of Prof. Johannes Broichhagen at the Leibniz FMP, known for pioneering innovative approaches to interrogate biological functions and the laboratory of Prof. David J. Hodson at the University of Birmingham, renowned for combining biophotonics, mouse genetics, modelling, and #chemical biology to address complex research questions in #metabolism.   • Addressing Morphine Side Effects and Opioid Drug Interactions with Fluorescence 💡   Recent research has shed light on the serious impact morphine can have on our brain's dopaminergic neurons. According to a recent study (https://lnkd.in/dbBVB3DB), morphine was found to significantly reduce neuron activity, disrupt mitochondrial function, and interfere with crucial signalling pathways. Morphine is often prescribed alongside other medications, especially for patients with complex treatment needs and along with other opioids (https://lnkd.in/dcyhPF6v) like hydrocodone and oxycodone, interacts with our body's enzymes—particularly the CYP450 essential for drug metabolism. To better detect harmful substances like morphine, scientists are developing advanced detection techniques. New fluorescent sensors are capable of detecting morphine in urine with incredible accuracy, at levels as low as 8 parts per billion (ppb)(https://lnkd.in/dx2VEjWE).   • Exploring Pharmacological Behavior in Morphine and Fentanyl Receptors Opioid receptors (ORs) a family of class A G-protein-coupled receptors (#GPCRs) have a significant importance in various pathophysiological processes in the Central Nervous System. Their study —specifically μ (MOR), δ (DOR), and κ (KOR)—is crucial in the scientific exploration of decision-making, addiction, and pain processing. #Morphine and #fentanyl are among the most used opioid drugs that confer analgesia and unwanted side effects through activation of G protein and arrestin signalling pathways of μ-opioid receptor. In recent years, driven by the #opioid abuse crisis, there has been a renewed interest in OR characterization. This has prompted researchers to search for new alternative ligands with diverse signalling properties that can further advance our understanding. #Fluorescent ligands have emerged as a promising alternative approach. When combined with computational simulations and biological assay, these tools can help characterize abnormal pharmacology, such as difficulties in fitting ligands into the binding pocket. This, in turn, allows for a deeper understanding of receptor binding and different binding modes. At Celtarys Research, in collaboration with MD Anderson Cancer Center, Texas University, we have designed #morphine fluorescent-labelled derivatives for use in in vitro assays, contributing to the ongoing efforts to better characterize opioid receptors and their interactions.
