[en] Chromatographically pure and stable anionic species of high charge density cations show higher uptake and give better melanoma images than monoclonal antibodies labeled with the same radionulcides. sup(99m)Tc-labeled antimelanoma antibody filtered for the removal of heavy colloids, hydroxides and oxides of 99m-technetium shows lower uptake in the liver and spleen. [en] Positron tomographs are often characterized by parameters like efficiency, resolution, reconstruction time, number of slices, etc. Due to differences in the method, additional information is needed when comparing conventional PET and TOFPET systems. Reconstruction gain, reduction of random coincidences, and the possibility of measuring cross slices are examined. Properties of two tomographic systems are compared and evaluated. (Author)

AbstractAbstract
[en] Commonly used methods for selecting specific monoclonal antibodies usually do not include parameters relevant for in vivo application, i.e. in immunoscintigraphy. We report on the criteria we used for screening a series of anti-melanoma antibodies. These included determination of immunoreactivity at different levels of radiolabeling. Antigen density was analyzed in various melanoma lines. In addition, the processing of antigen-antibody complexes was followed in metabolically active cells in order to find out whether a given antibody was anchored to the cell surface, internalized or shed. Furthermore, we studied conditions for enzymatic fragmentation of individual monoclonals as well as the immunoreactivity of labeled fragments, as antibody fragments gain increasing importance in immunoscintigraphy. AbstractAbstract
[en] A study of 125I-TSH pharmacokinetics in mice was performed in order to obtain more information on the peripheral tissues rich in thyrotropin receptors. By comparison with Na 125I-biodistribution, it was shown that 125I-TSH pharmacokinetics are quite different of that of free radioiodine. After 125I-TSH injection, an increased uptake is seen in thyroid, brain, eyes, pancreas and intestine and the pharmacokinetics of the radioactivity in these organs do not follow blood clearance. The results suggest thyrotropin receptors to be present in the above organs. (Author)
