AbstractAbstract
[en] High and selective accumulation in tumor cells is one important requirement for a BNCT agent. One way of increasing the tumor selectivity of BNCT agents may be the use of carbaborane-containing bisphosphonates. As some simple carbaboranylphosphonates have been proposed to accumulate in bone cells, it should be possible to improve the BNCT potential by varying esters of phosphonic acids. (author)

[en] Boron Neutron Capture Therapy (BNCT) was regarded as a potential cancer therapy but the successful application of BNCT to the treatment of cancer was still not approached due to their low water solubility, stability and low selectivity towards cancer cells. We developed a procedure for the synthesis water-soluble BNCT agents based on o-carboranyl-s-triazines containing polyol and polycarboxylic acid groups. (author)

AbstractAbstract
[en] A 39-year-old man had received Boron Neutron Capture Therapy (BNCT) in 1987 for a Grade II Astrocytoma. He gradually exacerbated and received a second operation in 1994. The mass taken in the second operation is almost competent with radiation necrosis. Following that, he shows no signs of recurrence. Currently, he has returned to full time employment in physical labor. This case suggests effectiveness of BNCT for rather low-grade astrocytomas. (author)

[en] In a previous study, we reported that sodium borocaptate (BSH) uptake was enhanced in combination with buthionine sulfoximine (BSO) in vitro. This report investigated the effect of the combination in vivo. In our results, the boron levels in blood and tissues were preserved highly in the BSO +BSH group, in comparison with the BSH only group. BSO has the potential to improve boron uptake in BNCT. [en] Lipids containing the closo-dodecaborate cluster have been developed. Two examples, S-(N,N-(2-dimyristoyloxyethyl) -acetamido)-thioundecahydro-closo-dodecaborate (2-) (B-6-14) and S-(N,N-(2-dipalmitoyloxyethyl) -acetamido)-thioundecahydro-closo-dodecaborate (2-) (B-6-16) have been prepared. With helper lipids, stable liposomes were obtained. (author)

[en] An accelerator-driven, thermal neutron beam facility, aimed at the Boron Neutron Capture plus Photodynamic Therapy (BNCT+PDT) experimental treatment of extended skin melanoma, is currently under construction at the INFN Laboratori Nazionali di Legnaro (LNL). New, boron loaded compounds have been at the purpose synthesized and a wide vitro and in vivo radiobiological investigation is under way. Special microdosimetric detectors have been developed and constructed at LNL, for properly on-line monitoring of all BNCT dose components and their qualities. Both microdosimetric and radiobiological measurements are being performed at the new, HYTHOR thermal irradiation facility at the ENEA-Casaccia TAPIRO fast reactor. (author)

[en] Two unnatural, boron-containing amino acids were synthesized by alkylation of S-(2-cyanoethyl)-thio-undecahydro-closo-dodecaborate(2-). S-(2-amino-2-carboxylpropyl)-thio-undecahydro-closo-dodecaborate (2-), containing a quaternary carbon atom, and O-(5-amino-5-carboxylpentyl)-oxy-undecahydro-closo-dodecaborate(2-) were evaluated for in vitro toxicity using V 79 Chinese hamster cells. [en] Three monoclonal antibodies against p-boronophenylalanine (anti-BPA MAb) were first prepared from the hybridoma by the modified reported method. The dissociation constants of anti-BPA MAbs against BPA and its related compounds were measured by using the enzyme-linked immunosorbent assay (ELISA). These anti-BPA MAbs had high binding affinity and specificity against BPA. (author)

[en] Magnetic resonance imaging (MRI) and boron-neutron capture therapy (BNCT) are quite attractive techniques for diagnosis and treatment of cancer, respectively. In order to develop the practical tools both for MRI and BNCT, the novel compounds containing both 19F and 10B atoms were designed and synthesized. In the present paper the syntheses of these compounds and their biological activity to tumor cells are reported. (author)
