AbstractAbstract
[en] Ablation of various regions of premigratory trunk neural crest which gives rise to the sympathetic trunks was used to remove sympathetic cardiac innervation. Neuronal uptake of [3H]-norepinephrine was used as an index of neuronal development in the chick atrium. Following ablation of neural crest over somites 10-15 or 15-20, uptake was significantly decreased in the atrium at 16 and 17 days of development. Ablation of neural crest over somites 5-10 and 20-25 caused no decrease in [3H]-norepinephrine uptake. Removal of neural crest over somites 5-25 or 10-20 caused approximately equal depletions of [3H]-norepinephrine uptake in the atrium. Cardiac norepinephrine concentration was significantly depressed following ablation of neural crest over somites 5-25 but not over somites 10-20. Light-microscopic and histofluorescent preparations confirmed the absence of sympathetic trunks in the region of the normal origin of the sympathetic cardiac nerves following neural crest ablation over somites 10-20. The neural tube and dorsal root ganglia were damaged in the area of the neural-crest ablation; however, all of these structures were normal cranial and caudal to the lesioned area. Development of most of the embryos as well as the morphology of all of the hearts was normal following the lesion. These results indicate that it is possible to produce sympathetically aneural hearts by neural-crest ablation; however, sympathetic cardiac nerves account for an insignificant amount of cardiac norepinephrine

AbstractAbstract
[en] The pressurised Metered Dose Inhaler (pMDI) has become the most prescribed drug delivery system for treating the respiratory diseases. However, the spray generation mechanism of these devices has not been extensively researched and there is very little information regarding the two-phase fluid dynamics associated with pre-atomisation inside the valve stem. The aim of the work presented in this paper is to provide high-quality, time-resolved imaging of the internal flow structures of pMDIs in an attempt to link the characteristics of the internal flow to external spray atomization processes. Visualisations of the aerosols in the near-orifice region findings from previous studies of commercial pMDIs and showed the following characteristics: (i) start-up transient (ii) fully developed spray with slow spray density variations and (iii) rapid spray density pulsations with large droplet production. The results clearly highlighted the potential of optical diagnostics in the development of improved accounts of the state of the flow inside a pMDI valve and its relationship with drop formation

AbstractAbstract
[en] Diabetes mellitus results from an absolute or relative deficiency of insulin producing pancreatic beta-cells. The adult human beta-cell's turnover rate remains unknown. We employed novel techniques to examine adult human islet beta-cell turnover and longevity in vivo. Subjects enrolled in NIH clinical trials received thymidine analogues [iododeoxyuridine (IdU) or bromodeoxyuridine (BrdU)] 8-days to 4-years prior to death. Archival autopsy samples from ten patients (aged 17-74 years) were employed to assess beta-cell turnover by scoring nuclear analog labeling within insulin staining cells. Human adult beta-cell longevity was determined by estimating the cells genomic DNA integration of atmospheric carbon-14 (14C). DNA was purified from pancreatic islets isolated from cadaveric donors; whole islet prep DNA was obtained from a 15 year old donor, and purified beta-cell DNA was obtained from two donors (age 48 and 80 years). 14C levels were then determined using accelerator mass spectrometry (AMS). Cellular 'birth date' was determined by comparing the subject's DNA 14C content relative to a well-established 14C atmospheric prevalence curve. In the two subjects less than age 20 years, 1-2% of the beta-cell nuclei co-stained for BrdU/IdU. No beta-cell nuclei co-stained in the eight patients more than 30 years old. Consistent with the BrdU/IdU turnover data, beta-cell DNA 14C content indicated the cells 'birth date' occurred within the subject's first 30 years of life. Under typical circumstances, adult human beta-cells and their cellular precursors are established by young adulthood.

AbstractAbstract
[en] The dosimetric characteristics of a production pilot multi-leaf collimator (Elekta Beam ModulatorElekta Oncology Systems, Crawley, UK) having a 4 mm leaf width (at isocentre) have been investigated. Characteristics explored included leaf bank set-up, penumbra width (80-20%) as a function of leaf position, leaf positioning reproducibility, interleaf leakage and leaf transmission. The penumbra values for leaf ends were measured to be between 4.2 and 4.8 mm for various large rectangular fields studied using Kodak X-omat V film at isocentre (1.5 cm deep). Similar films were taken with a standard 1 cm width multi-leaf collimator (MLC) and the penumbra for leaf ends was found to range from 4.3 to 5.2 mm. Other results showed that the rounded leaf tip provided tight control of the penumbra across the leaves' full range of travel. The positioning of the leaves was within a 0.5 mm range when approaching from the same direction. The maximum interleaf leakage was found to be 1.7% and the average leaf transmission less than 1.0%. No major differences were observed in leakage and transmission with changing gantry angle

AbstractAbstract
[en] The Intensity Frontier refers to a diverse set of particle physics experiments using high- intensity beams. In this paper I will focus the discussion on the computing requirements and solutions of a set of neutrino and muon experiments in progress or planned to take place at the Fermi National Accelerator Laboratory located near Chicago, Illinois. The experiments face unique challenges, but also have overlapping computational needs. In principle, by exploiting the commonality and utilizing centralized computing tools and resources, requirements can be satisfied efficiently and scientists of individual experiments can focus more on the science and less on the development of tools and infrastructure. AbstractAbstract
[en] We present the development and validation of the Higgs Optimized b Identification Tagger (HOBIT), a multivariate b-jet identification algorithm optimized for Higgs boson searches at the CDF experiment at the Fermilab Tevatron. At collider experiments, b taggers allow one to distinguish particle jets containing B hadrons from other jets; these algorithms have been used for many years with great success at CDF. HOBIT has been designed specifically for use in searches for light Higgs bosons decaying via H→bb¯. This fact combined with the extent to which HOBIT synthesizes and extends the best ideas of previous taggers makes HOBIT unique among CDF b-tagging algorithms. Employing feed-forward neural network architectures, HOBIT provides an output value ranging from approximately −1 (“light-jet like”) to 1 (“b-jet like”); this continuous output value has been tuned to provide maximum sensitivity in light Higgs boson search analyses. When tuned to the equivalent light jet rejection rate, HOBIT tags 54% of b jets in Monte Carlo simulated Higgs boson events (mH=120 GeV/c2) compared to 39% for SecVtx, the most commonly used b tagger at CDF. We present features of the tagger as well as its characterization in the form of b-jet finding efficiencies and false (light-jet) tag rates.

AbstractAbstract
[en] A workshop was held at Fermilab on March 20-21, 2013 to discuss the development of liquid argon time projection chambers (LArTPCs) in the United States. The workshop was organized under the auspices of the Coordinating Panel for Advanced Detectors, a body that was initiated by the American Physical Society Division of Particles and Fields. All presentations at the workshop were made in seven topical plenary sessions: i) Argon Purity, ii) Cryogenics, iii) TPC and High Voltage, iv) Electronics, Data Acquisition and Triggering, v) Scintillation Light Detection, vi) Calibration and Test Beams, and vii) Software. This document summarizes the current efforts in each of these areas. It also highlights areas in LArTPC research and development that are common between neutrino experiments and dark matter experiments
