AbstractAbstract
[en] The invention refers to a method and an arrangement for influencing and guiding a high-energy particle beam accelerated in a cyclotron. The aim is to generate a focussed particle beam at the direction of emission from the cyclotron by a large compensation of the fringing magnetic field effects of the cyclotron magnet. The arrangement has an electrostatic quadrupole singlet at the cyclotron-sided end of the beam guiding system. The axis of the escaping particle beam corresponds to the axis of the quadrupole singlet approximately. It is favourable that the quadrupole singlet electrodes have a bend adapted to the beam axis

The design procedure and the construction of the electromagnet, the vacuum chamber with monant, the vacuum system, the power supply and the detecting system are demonstrated. AbstractAbstract
[en] Advantages of a stacked assembly of target support components for 64Cu production via 64Ni(p,n)64Cu reaction were reported recently. The present work shows the applicability of these composite targets for beam currents up to 22 μA. Gold and platinum foils were evaluated as 64Ni backing. The effective specific activity (ESA) and specific activity (SA) were determined by TETA titration at room temperature and at 80 °C and compared with additional copper quantification results via ICP-MS and stripping voltammetric trace analysis (VA).

AbstractAbstract
[en] Aim: characterisation of the influence of different polymeric tube materials of a water target system, used for the production of 18F activity, on the specific activity of radiotracers. Material, methods: target water samples taken from different locations of the 18F water target system of a Cyclone registered 18/9 cyclotron, equipped with Teflon registered (PTFE) or polypropylene (PP) tubes, were analyzed for non-radioactive [19F]fluoride content. [19F]Fluoride content was measured by ion chromatography (IC20, Dinoex) with suppressed conductivity detection. Both the ion chromatographic results and the amount of 18F activity produced were used for the calculation of the specific activity (SA) of [18F]fluoride at the start of the labelling synthesis. To check these results, the SA of the labelled receptor ligand [18F]ZK811460 was also determined by using the different tubing materials. Results: dose-exposed PTFE tubes of the target dispensing (loading) system were identified to be a major source of [19F]fluoride contamination. Conclusion: by replacing PTFE tubes of the target dispensing system with PP tubes, the content of 19F was reduced considerably resulting in an increase of SA of the radiotracer [18F]ZK811460 by factor of two. AbstractAbstract
[en] 64Cu (t1/2 = 12.7 h; β+ 0.653 MeV, 17.4%; β-: 0.578 MeV, 39%) is a radionuclide of high interest for nuclear medicine. 64Cu has already been utilized for research purposes due to its favorable decay characteristics and its ability to be produced at high specific activity. Bispidines and azamacrocycles containing pyridine pendant arms form thermodynamically stable complexes with copper (II). These chelating agents possessing an additional linker group provide an attractive lead for labeling biomolecules with copper radionuclides. Experimental: First experiments to adapt the described 64Cu-production process at a PET cyclotron CYCLONE 18/9 were performed. 10 MeV protons and a proton beam current of 10 μA for 15 to 60 min were used. The yields of the 64Ni(p, n) 64Cu nuclear reaction were comparable to reference values [179-331 MBq/μA/h versus 185-248 MBq/μA/h]. Two novel ligands with carboxylic groups a hexadentate bispidine 1 and a derivative of 1,4,7-triazacyclononane (TACN) 2 were prepared. Labeling experiments of these ligands with 64Cu2+ indicated the rapid formation of stable 64Cu-complexes under mild conditions. The bioconjugates 3 and 4 were obtained by coupling the bombesin (BN) derivate βAla-βAla- [Cha13, Nle14] BN (7-14) to the ligands 1 and 2 (HBTU, DIPEA, DMF, r.t.). Results and Discussion: The bioconjugation of 1 and 2 to a bombesin derivative was successfully achieved via amide formation to give 3 and 4. The free ligands and the bioconjugates were labeled with 64Cu2+ and the resulting complexes were found to be stable in the presence of a large excess of competing ligands such as cyclam and even in rat plasma. Conclusion: The bispidine ligand 1, the TACN derivative 2 and their bombes in bioconjugates 3 and 4 are able to form complexes with high stability. These bifunctional chelating agents may be attractive candidates for the development of new copper radiopharmaceuticals for diagnosis (64Cu) and therapy (67Cu). Preliminary studies on the bio-distribution of 64Cu-complexes of 3 and 4 are currently underway
