AbstractAbstract
[en] In order to solve the conjugate heat transfer problem formed by the guide tube of sodium-cooled fast reactor hydraulic suspended passive shutdown mechanism and the liquid sodium on its inner and outer sides, Trelis, a pre-processing software was used to generate the global high-quality hexahedral mesh, and a solver called chtMultiRegionSimpleFoam of OpenFOAM, which is an open source CFD software, was applied. The temperature distribution results of the numerical analysis of the guide tube were obtained. The results show that the temperature difference in the range of 0.07 m in the circumferential direction of the inner wall of the guide tube is 97℃, and the temperature difference in the range of 0.17 m in the axial direction is 80℃. In such a narrow range, the temperature difference will generate large thermal stress, and the numerical analysis results of which will provide an important input for the mechanical design of the guide tube. [en] Highlights: • Identification of particular causes of failure for common cause failure analysis. • Comparison two formalisms (GO-FLOW and Discrete Time Bayesian network) and establish the correlation between them. • Mapping the GO-FLOW model into Bayesian network model. • Calculated GO-FLOW model with common cause failures based on DTBN. - Abstract: The GO-FLOW methodology is a success-oriented system reliability modelling technique for multi-phase missions involving complex time-dependent, multi-state and common cause failure (CCF) features. However, the analysis algorithm cannot easily handle the multiple shared signals and CCFs. In addition, the simulative algorithm is time consuming when vast multi-state components exist in the model, and the multiple time points of phased mission problems increases the difficulty of the analysis method. In this paper, the Discrete Time Bayesian Network (DTBN) and the GO-FLOW methodology are integrated by the unified mapping rules. Based on these rules, the multi operators can be mapped into DTBN followed by, a complete GO-FLOW model with complex characteristics (e.g. phased mission, multi-state, and CCF) can be converted to the isomorphic DTBN and easily analyzed by utilizing the DTBN. With mature algorithms and tools, the multi-phase mission reliability parameter can be efficiently obtained via the proposed approach without considering the shared signals and the various complex logic operation. Meanwhile, CCF can also arise in the computing process.

AbstractAbstract
[en] Accumulating evidence revealed the abnormal expression of KLF5 in human cancers while its role in melanoma remains uncharacterized. This study aimed to explore the role of KLF5 in the proliferation and metastasis of melanoma. Bioinformatics analysis was performed to detect WWP1, BAP1 and KLF5 expression in melanoma, followed by expression determination on clinical tissues from melanoma patients and cancer cells. The cancer cells were infected with lentivirus expressing KLF5 or BAP1 while PI3K, AKT and mTOR expression was detected and autophagy was observed. Treated cells were injected to mice when tumor growth was measured and autophagy-related protein was detected. Plasmids expressing WWP1 and Ub-K48 were co-transfected into treated melanoma cells while immunoprecipitation assay was performed to determine the interaction among KLF5, WWP1, and BAP1. WWP1 was poorly expressed in melanoma cells and tissues whereas KLF5 was highly expressed and was positively correlated to poor prognosis. KLF5 promoted melanoma cell malignant phenotypes as well as inhibited autophagy. Interestingly, KLF5 contributed to activation of PI3K-AKT-mTOR signaling pathway, thereby inhibiting autophagy in melanoma cells. WWP1 mediated K48-linked ubiquitination of KLF5 to promote its degradation, and BAP1 antagonized this modification and stabilized KLF5 protein expression. Besides, BAP1 promoted KLF5-mediated growth of melanoma in vivo. Taken altogether, BAP1 antagonized WWP1-mediated ubiquitination of KLF5 to inhibit autophagy and promote melanoma development.

AbstractAbstract
[en] Accurate remaining useful life (RUL) prediction for critical components of nuclear power equipment is an important way to realize aging management of nuclear power equipment. The electric gate valve is one of the most safety-critical and widely distributed mechanical equipment in nuclear power installations. However, the electric gate valve's extended service in nuclear installations causes aging and degradation induced by crack propagation and leakages. Hence, it is necessary to develop a robust RUL prediction method to evaluate its operating state. Although the particle filter(PF) algorithm and its variants can deal with this nonlinear problem effectively, they suffer from severe particle degeneracy and depletion, which leads to its sub-optimal performance. In this study, we combined the whale algorithm with regularized particle filtering(RPF) to rationalize the particle distribution before resampling, so as to solve the problem of particle degradation, and for valve RUL prediction. The valve's crack propagation is studied using the RPF approach, which takes the Paris Law as a condition function. The crack growth is observed and updated using the root-mean-square (RMS) signal collected from the acoustic emission sensor. At the same time, the proposed method is compared with other optimization algorithms, such as particle swarm optimization algorithm, and verified by the realistic valve aging experimental data. The conclusion shows that the proposed method can effectively predict and analyze the typical valve degradation patterns
