AbstractAbstract
[en] A field experiment has been conducted to increase internal CO2 and Rubisco activity detected by 14C and to determinate which factors influence this activities. Plant material used was cotton plants which internal CO2 concentrations and Rubisco activity was observed at 35, 50, 65, 80 days after planting (DAP). Treatments applied were methanol with concentrations of 0%, 10%,20% and 30% at available water (AW) at 25-50% AW, 50-75% AW, 75-100% AW. Results obtained showed that application of methanol at concentration of 20% at 75-100% AW, increase internal CO2 from 266.60 ppm to 295.10 ppm (11 % increase) and this will also increase Rubisco activity from 3.81 to 14.28 (μmol. CO2 menit-1 (μmol. Rubisco-1). This increase is expected to push photosynthesis rate and result in increase cotton yield. The use of 14C was satisfactorily detected the amount of carbon. (author)

AbstractAbstract
[en] Two experiments (dry and wet seasons) have been conducted to study effect of Azolla inoculated in lowland rice culture. Five treatments combination of urea and Azolla tested were: N-0, N-0+Az, N-45+Az, N-90+Az, and N-90. Experiments were designed with completely randomized design (CRD) and each treatment was replicated 4 times. 15N-labelled urea was used in this experiment to determine N-derived from fertilizer. Results of this experiments showed that planting season did not influenced production of grain, total-N percentage, and total-N uptake of the plant. Azolla inoculation was able to increase dry matter production, percentage of total-N, and plant total-N uptake up to 36 %. The fertilizer use efficiencies were increased 16 % in dry season and 23 % in wet season respectively. This condition expressed that Azolla inoculation in lowland rice could present N-logg through vaporization and water current. (author)

AbstractAbstract
[en] Five dairy cattle, cattle A: 6th parity; cattle B: 7th parity; cattle C: 2nd parity; cattle D and F: 3rd parity, were used in this study. According to Health Extension and Artificial Insemination Technicians anamneses and according to farmers who own the animals, these cattle were showing reproductive failure, and RIA technique was used to study the symptoms. For this purpose, milk progesterone sample were collected twice a week for five weeks to follow the biological reproductive status of every animal. Result from the analysis were plotted for each individual animal and shows that cattle A and B were acyclic, which indicated that no reproductive activity post partum have occurred in both animals; cattle C in the status of prolonged oestrus cycle post partum; and cattle D and F were in the status of recovery of oestrus cycle post partum. With the availability of historical record of the cattle and confirmation of biological status by Health Technicians, the reproductive disorder, which leads to the failure of AI in dairy cattle, can be monitored by RIA Progesterone technique. AbstractAbstract
[en] Pumpkin powder (Cucurbita pepo L.) can be used as food diversification originally derived from plant as local product and raw material for making jam, cookies, spaghetti and instant noodles. Effects of gamma irradiation on pumpkin powder at medium doses during storage was observed. Whole pumpkin was cut into small pieces, then blanched at 90 oC for 5 min. followed by transforming it into paste form using wet grinder. The pumpkin paste was dried using drum drier then powdered by willey mill until a 60 mesh particle size was obtained. The powder was packed in a high density polyethylene (HDPE) pouch at 0.01 mm thickness. Pumpkin powder was irradiated at doses of 3 and 5 kGy respectively then stored at 28-30 oC for 2 and 4 months, respectively. Unirradiated individual pouch was also used as control. Irradiation with doses of 3 dan 5 kGy could reduce 2 log cycle of total bacterial count, no mold nor yeast growth in irradiated samples. Irradiation with doses of 3 and 5 kGy could effectively reduce the microbiological counts of the pumpkin powder observed. Pumpkin powder originated from 2 different habitat i.e., Cipanas and Brebes did not show any differences in some physico-chemical characteristics such as moisture content, Aw, protein, fat, starch, amylose contents, colour, but significantly different in sugar content. Both radiation and storage treatments could significantly reduce some of the physico-chemical characteristics of the investigated samples, but did not affect the general quality of pumpkin powder. AbstractAbstract
[en] Study about effects of water content levels on yield components of peanut mutant lines has been conducted. Mutant lines tested were A20/3/PsJ, B30/12/10 and D25/3/2. The original varieties/lines of the mutants were, AH1781Si, Kidang and Pelanduk was included as control. The treatments for levels of water application were 100%, 75%, 50% and 25%, where 100% water available means a difference between water content at field capacity and at content in permanent wilting point. Based on the data of parameters observed, to interaction between varieties/lines and water content levels was not found except for dry matter weight. For this parameter, the highest dry matter was observed when B30/12/10 mutant line was watered by 100% water available. Nevertheless, based on relative values of number of mature pod, dry pod weight and 50 nut weight the A20/3/PsJ mutant line seems tolerant to drought. (author)

AbstractAbstract
[en] Atom ita-4 rice variety was crossed with IR-64 variety in the greenhouse at the Center for Application of Isotopes and Radiation-Batan, Pasar jumat in the wet season of 1994/1995. F1 Seeds derived from Atomita-4/IR-64 crossing were irradiated by gamma rays at of 0.2 kGy dose. F1 seeds were grown to obtain F2 M2 seed, and then selection of pedigree were carried out at F2 generation. Six mutants lines were obtained purified and screened on biotypes 1, 2 and 3 brown plant hopper and bacterial leaf blight resistance by IRRI standard screening methods. The six mutant lines were tested for their potential yield at Pusakanegara and then continued tested in yield multi location test at several locations in Indonesia. Results of the screening test to brown plant hopper showed that two mutant lines Obs-1653/PsJ and Obs-1656/PsJ were resistant to biotype 1, biotype 2 and medium resistant to biotype 3. Obs-1653/PsJ and Obs-1656/PsJ also showed resistance to bacterial leaf blight strain 3 and medium resistance to strain 4. Results in the yield multi location test showed that Obs-1653/PsJ and Obs-1656/PsJ have highest yielding potential compared to IR-64 and Memberamo varieties. Those two mutant lines were released as new varieties under the name Merauke and Kahayan in 2001 and 2003 respectively. (author)
