Important News – Incompatible Analgesic containing MCT Oil We have found that Ethiqa XR long acting analgesia which contains MCT oil is not compatible with iPRECIO pumps. The use of Ethiqa XR as analgesic has been found to weaken iPRECIO pump reservoir and cause leaks. More significant damage has also occurred. We recommend not to use Ethiqa XR or any analgesic formulations containing oils. If the use of Ethiqa XR is important, please contact Primetech Corporation for further information. Finally, as a reminder, iPRECIO Pumps are not compatible with oils as vehicle and oils will weaken and will destroy the reservoir and catheter. " Our Mission with iPRECIO® Product Portfolio: Make it as easy as possible to evaluate efficacy of molecules/agents in different animal models and species. Make the results as relevant as possible to the clinic. (Exposure profile, natural dose patterns, quantitative pharmacology, untethered free moving (stress free, group housing (socialization), ..…) The use of iPRECIO® pumps will have a positive impact on experimental conditions and the relevance of results achieved. Initial costs maybe higher but in the long run the use of the pump could be cost saving in the context of the whole compound development.

Pump Programming: iPRECIO SMP-200 micro-infusion pumps were implanted in female Sprague Dawley rats at postnatal day 60 (P60). After a one-week recovery period post-implantation, pumps delivered morphine or saline in a scheduled regimen: Initial dosing of 10 mg/kg/day in three doses, incrementally increasing weekly by 2 mg/kg/day, up to 16 mg/kg/day. On approximately embryonic day 18 (E18), dosing frequency was reduced to twice daily, mitigating respiratory depression in pups observed in earlier experiments. 🧑🔬💊 We need to talk more biopharma R&D productivity this year 📊💡 My take on the fresh "special collection" on this topic from McKinsey & Company 📍 Big picture is spot on: Biopharma's innovation challenges remain, and return on investment on drug R&D continues to be under pressure 📍 The obvious: McK putting out a "special collection" of 100+ pages on R&D productivity at the start of 2025 just shows that they know biopharma is struggling to tackle these challenges 🧩 The R&D productivity equation is a composite of 5 levers: 👉 Pipeline volume 📍 The total, global pipeline is growing - trend: ↗️ (at least there is volume in the pipelines...great, as long as there is QUALITY, too) 👉 Probability of success 📍 PoS has not improved meaningfully over several decades - trend: ➡️ (McK puts it as ↘️, so I am a bit less pessimistic on this one; again, depends somewhat on dataset) 👉 Value per drug 📍 Peak sales per asset is under pressure, but let's go with McK here and say it is stable - trend: ➡️ 📍 Note that biopharma is increasingly relying on the "blockbuster model" (big wins make the day and make the wheels go round to pay for all your pipeline attrition and for the significant fraction of your drug launches that ends up with a negative ROI) 👉 R&D cost per drug 📍 Not getting cheaper to do drug R&D - trend: ↗️ (pharma R&D is now in the range of $250 billion per year) 👉 R&D speed (cycle times across R&D stages) 📍 McK states speed is increasing, but - again depends on dataset - I think this is too optimistic, so I would not put trend better than flat: ➡️ 🤕 ...and across the industry, these key levers are moving in "opposite" direction with the effect that positive and negative movements in the levers *cancel out*, muting any substantial and meaningful uptick in R&D productivity (it is like the industry is trying to address some levers, but then give away all the improvement to other levers) 📍 Where biopharma stands on R&D productivity today: Following decades of decline, we saw - looking at some metrics - signals from ~2010 that R&D productivity was starting to switch direction; however, the uptick has been erratic - with no convincing evidence that the industry is really on the right track Two other insights to take away: 🏃♀️➡️ Pharma pipelines are increasingly chasing the same targets - not a new phenomenon, but a reminder that no easy "blue ocean" strategy exists in this business (good luck competing in "red ocean") 🌀 Biopharma pipelines are extremely dynamic with an annual turnover of 20+% due to program discontinuation - it's a formidable task to replenish a fifth of your pipeline ...
