My posts over the past couple of weeks sparked some lively debates, with a few people thinking I was advocating for a "bigger is better" approach to bioreactor scaling. While CAPEX and OPEX are crucial considerations, they’re far from the only factors when determining the optimal size for a production-scale bioreactor. In fact, I published papers on this topic five years ago, including a graphic representation of typical bioreactor sizes and the types of products they’re best suited for (see below). Market demand, process yields, regulatory requirements, and time-to-market all play key roles in optimizing reactor scale. This is why a Techno-Economic Analysis (#TEA) is so important—it balances both technical and economic factors to ensure #scalability and #profitability. Recently, a client approached us with a puzzling observation during a 480m³ stirred tank #bioreactor run. As they increased the liquid level during a fed-batch #fermentation - while keeping airflow and agitation constant - their mass transfer seemed to improve! Strange, right? According to all the empirical correlations, increasing liquid volume should reduce mass transfer, as both P/V (power per unit volume) and VVM (volume of air per unit volume of liquid per minute) decrease. After diving deeper with #CFD (Computational Fluid Dynamics) analysis, we uncovered the root cause: the reactor was operating in the partially dispersed regime - agitation speeds were too low for the given airflow. In this regime, gas dispersion only occurred efficiently above the second impeller. As the liquid volume above the impeller increased, so did the overall gas holdup, driving up the #kLa (mass transfer coefficient). This case serves as a powerful reminder: when designing or operating bioreactors based on empirical correlations, always ensure that you understand the applicable flow regime. Most correlations assume fully dispersed conditions, which may not always be the case. If you have been faced with challenges like this, feel free to reach out.
