Always good to see the hard work by students gets to a point that bring recognition for them. My heart-felt congratulations to Ivan (former graduate of MS Course in University of Manchester in Modelling) who managed to get this piece to finish line with the help of several top class scientists at the Certara Predictive Technology arm. 📢 New #CAPKR paper ALERT! We are pleased to share our most recent publication "Absolute Membrane Protein Abundance of P-gp, BCRP and MRPs in Term Human Placenta Tissue and Commonly Used Cell Systems: Application in PBPK Modeling of Placental Drug Disposition" in the journal Drug Metabolism and Disposition (DMD) 💡 This study quantified the expression of key transporters in the human placenta and various placental cell lines, revealing significant expression variations. 💡 By integrating with PBPK modeling, the study highlighted the importance of transporter abundance data in understanding and predicting placental drug disposition. 💡 This study offers a promising tool to investigate maternal, placental, and fetal drug exposure. Excited to share our accepted paper that establishes a translational physiologically based pharmacokinetic modelling framework for human aldehyde oxidase in #PSPJournal, led by Nihan Izat along with collaboration with industry members of Centre for Applied Pharmacokinetic Research (CAPKR): https://lnkd.in/ecEGWzwa Although increasing numbers of investigational drugs are substrate for aldehyde oxidase (e.g., kinase inhibitors), there are gaps in the clinical pharmacology toolbox that create uncertainty and risk for bringing these through clinical development: ⚠️ Lack of selective and potent in vitro inhibitors for estimation of fraction metabolised by AO ⚠️ Uncertainty in the contribution of extra-hepatic AO ⚠️ No clinical index inhibitors to evaluate drug-drug interaction risk Our research found that: 💡 Clinical mass balance data, supported by robust reaction phenotyping, is crucial to enable estimation of fraction metabolised by AO within PBPK models 💡 For dual AO-CYP substrates, CYP DDI data can be applied for indirect validation of fraction metabolised by AO 💡 Extrahepatic AO will, in general, have limited contribution to systemic clearance (exception for high extraction AO substrates, where renal AO becomes relevant) #ASCPTJournalFamily #PBPK #CAPKR #research #collaboration
