Determining sensitivity is based on the toxicity of the product manufactured. The most toxic/ active component in the product must be removed to a level at which any residue would not impact the subsequent product manufactured in the equipment cleaning. The acceptable level calculation of the amount that might be potentially carried into the next manufacturing batch. The calculation process is based on the target toxicity, equipment total surface area, surface area swabbed, the surface material [SS, nylon, PP, Teflon, etc.], swab recovery on each surface and the limit in the next product. In my experience, the choice of a method always depends on the analyte. If the goal is to identify multiple analytes from a single sample, specificity outweighs sensitivity. In such cases, users should opt for sophisticated techniques like HPLC or LC/MS. However, if the aim is to identify a single analyte, users should consider methods that provide an overall load of residues, such as TOC and conductivity. These methods are sensitive enough to determine the overall load and can be used in dynamic environments with in-line sensors. Regardless, both sensitivity and specificity play a crucial role in the cleaning validation process
