Do you know what a chromophore is❓ 👉The term chromophore defines a group of atoms capable of giving color to a substance. More specifically, a chromophore represents an atom or group of atoms of a molecular entity responsible for the onset of a given spectral band following an electronic transition. The main chromophores of laser therapy are hemoglobin, hemoglobin oxide, water, melanin and cytochrome c oxidase. The main chromophore of photobiomodulation is the cytochrome c oxidase. The cytochrome c complex, found in the inner membrane of cellular mitochondria, is a vital component of the electron transport chain that drives cellular metabolism. When laser light is absorbed, cytochrome C is stimulated, leading to an increase in the production of adenosine triphosphate (ATP), the molecule that facilitates the transfer of energy within the cell. The luminous energy of laser light, interacting with biological tissues, is transformed into chemical energy (Photochemical Effect), mechanical energy (Photomechanical Effect) and thermal energy (Photothermic Effect). 𝗟𝗮𝘀𝗲𝗿 𝗧𝗵𝗲𝗿𝗮𝗽𝘆 with 𝗣𝗵𝗼𝘁𝗼𝗯𝗶𝗼𝗺𝗼𝗱𝘂𝗹𝗮𝘁𝗶𝗼𝗻 (PBM) is indicated to reduce pain associated with various pathologies and applied using protocols that work according to the length of irradiation and the therapeutic dose. Laser therapy has a direct impact on pain transmission by reducing the response of nociceptors. In this study, the effects of two specific wavelengths were examined: 800 and 970 nm, widely applied in the clinical setting and known for important analgesic effects. The results highlight how mitochondria are the main target and how it is the reduction of adenosine triphosphate. In particular, the 800 nm wavelength induced mitochondrial dysregulation. When the DRG neurons were first illuminated by the different laser treatments and then stimulated, only the 970 nm wavelength reduced the calcium response, both in amplitude and in frequency.
