ISS is committed to the development and design of highly sensitive fluorescence instrumentation for scientific research, clinical and industrial applications. A subsidiary of ISS, ISS Medical, markets non- invasive near-infrared instrumentation for absolute measurements of oxygen and hemoglobin concentrations in tissue (brain and muscle) and functional brain imaging. ISS has a long history of innovation beginning with time-resolved fluorometers with picosecond resolution, we manufacture fluorescence instrumentation for time-resolved (frequency-domain and time-domain) and steady-state measurements. Including, a line of modular components for fluorometers: laser diodes, LEDs, high pressure cell and fiber optic sensors amongst an extensive line of accessories. Our microscopy line provides single molecule tracking and nanoimaging. Our Alba combines a scanning confocal lifetime imaging microscope with fluorescence correlation spectroscopy and we also provide components for microscopy: data acquisition cards for FCS and FLIM, laser launchers, galvo-scanning mirrors and detector units.

Phasor Plots & VistaVision Part 3 Results & Discussions: The Phasor Plots Analysis of Label-Free NADH Lifetime Images Phasor plot analysis in fluorescence lifetime imaging microscopy (FLIM) of label-free NADH provides critical insights into human disease processes by leveraging endogenous autofluorescent molecules like NADH and FAD. FastFLIM, combined with phasor plots, simplifies NADH imaging in live cells, tissues, and organisms by differentiating between free and protein-bound forms of NADH—each with similar spectra but distinct lifetimes. Typically, this involves complex double-exponential decay fitting; however, the Multi-Image Phasor Analysis (MIPA) module simplifies this by enabling single-lifetime analysis of NADH forms. Through this method, researchers can directly quantify the free vs. bound NADH states within cells. The MIPA software provides phasor coordinates, fractions, and pre-exponential factors for both NADH forms, along with intensity- and amplitude-weighted lifetimes at each pixel. Phasor Plots & VistaVision Part 3 Results & Discussions: Improvements of Image Quality and Quantification by TR-Unmixing Time-resolved (TR) imaging enhances both image quality and quantification by incorporating ultrafast timing information, captured at the fluorescence lifetime scale, into traditional intensity-based imaging. TR-unmixing on phasor plots significantly improves signal clarity by separating contributions from scatter and white noise. These noise sources, appearing at distinct phasor coordinates (zero and infinite lifetimes), can be easily filtered out. This method is particularly valuable in fluorescence microscopy where signals from fluorescent dyes overlap with autofluorescence, complicating quantitative analysis. For example (Figure 1), Acridonylalanine (ACD), a fluorescent amino acid, has a spectral overlap with NADH, an endogenous autofluorescent molecule, making ACD quantification challenging. However, TR-unmixing effectively separates ACD (with a longer ~15 ns lifetime) from NADH (lifetime < 5 ns), resulting in distinct images for accurate signal quantification.
