🚀New Research in Cancer Cell on GBM: The Most Aggressive Adult Brain Tumor🧠 🧐 In this study, researchers hypothesized that daily signaling in the body regulates GBM growth and synchronizes its circadian rhythms. They found that glucocorticoids, via glucocorticoid receptor (GR) signaling, either promote or suppress GBM growth, influenced by the clock genes Bmal1 and Cry. Blocking circadian signals, such as vasoactive intestinal peptide or glucocorticoids, can slow down GBM growth. Data from The Cancer Genome Atlas (TCGA) indicates that high GR expression increases the risk of mortality. 🌟 New Research on Nonalcoholic Fatty Liver Disease (NAFLD) 🌟 👏 A recent study published in Cell Metabolism explores the role of the deubiquitinating enzyme RPN11 in NAFLD and nonalcoholic steatohepatitis (NASH). The authors found that hepatocyte-specific knockout of RPN11 protects against diet-induced liver steatosis, insulin resistance, and steatohepatitis. ✨ Key Findings: ✅ RPN11 stabilizes METTL3, enhancing the m6A modification of ACSS3, which in turn upregulates lipid metabolism genes through histone propionylation. ✅ The RPN11-METTL3-ACSS3 pathway is activated in the livers of NAFLD patients. ✅ Pharmacological inhibition of RPN11 using Capzimin effectively ameliorates NAFLD and related metabolic disorders in mouse models and reduces lipid accumulation in human hepatocytes. ✅ This study highlights RPN11 as a novel regulator of NAFLD/NASH and suggests its inhibition as a potential therapeutic strategy. New research published in Signal Transduct Target Ther uncovers a promising therapy for anemia. 🥳 🥳 🥳 Erythropoiesis, vital for blood formation, is often disrupted by diseases, leading to anemia and blood shortages. Traditional treatments like EPO often fail, especially for hereditary anemias like DBA. The study found that BRAF inhibitors (BRAFi), used for melanoma, can promote progenitor cell proliferation by delaying erythroid differentiation. Effective in cytokine-restricted conditions and DBA samples, BRAFi enhances MAPK cascade in wild-type cells. In vivo studies show BRAFi can improve hematopoiesis and treat anemia, including DBA.
