💉 How does LNP size affect vaccine performance? 👩🔬 In "Impact of lipid nanoparticle size on mRNA vaccine immunogenicity”, Kimberly Hassett and colleagues from the Moderna team took a systematic approach to modifying the particle size of #mRNA-#LNPs #vaccine formulations encoding for a cytomegalovirus surface pentamer complex and for envelope glycoprotein gB by adjusting formulation process variables while keeping lipid composition constant. 🔬 #CryoEM nicely confirmed the intended increase in particle size as measured by NTA and DLS. Furthermore, cryo-EM images showed a wider size range as well as the presence of #multilamellar structures in the larger particle formulations. The particle surface was found to be less polar with increasing size, and after showing that total mRNA encapsulation did not correlate with LNP size measurements, the impact of LNP size on innate immunogenicity in mice and non-human primates was studied. 🧪 The researchers show that in mice LNPs smaller than 85–100 nm induced significantly lower immunogenicity with respect to the size interval of 100–200 nm, whereas in non-human primates all tested LNPs in the 60-150nm size range triggered a strong immune response when the LNPs were delivered intramuscularly, suggesting species-specific differences in LNP size sensitivity. 📑 Read the study: https://lnkd.in/gu4fgxTs 👉 Learn more about phase appropriate characterization of #lipidnanoparticles: https://lnkd.in/g5s7Pk7z

🗑️ The N- and C-degron pathways are essential processes in protein quality control, where E3 ligases identify specific sequences at the protein's ends to facilitate targeted degradation. The selectivity of the ubiquitin-proteasome system depends on specific interactions between E3 ligases and their substrates. 🔬 In the study "Structural Basis for C-degron Selectivity across KLHDCX Family E3 Ubiquitin Ligases," Scott et al. use #cryoEM, in combination with X-ray crystallography, to uncover key principles of C-degron recognition in the KLHDCX family of Cullin-RING ligases (CRLs). The structures reveal that these E3 ligases employ a common C-terminal anchor motif (R/F-S-R), which appears in distinct spatial locations, creating unique molecular environments that enable distinct specificities. 💡 This research provides valuable insights into how this family of #E3 #ligases contributes to the broader biological mechanisms of protein quality control.
