#Fluorescence #Microscopy without filters? A new article appearing in Science Advances introduces a #DeepLearning-based approach to avoid the need for optical filters with fluorescence imaging. The described microscope design uses angled illumination with a tunable LED (independent of the detection path) and a color camera for detection. This method is demonstrated for isolating fluorescence signal, even with a significant contribution by scattered excitation light, and with imaging channels across the visible spectrum. INSIHGT is a new method for enhancing the depth penetration of antibodies and other macromolecular complexes for more homogenous labeling, paving the way for more impactful #SpatialBiology experiments in centimeter-scale tissue. This method works in two parts, first formaldehyde-fixed tissue is incubated with antibody and a weakly coordinated superchaotrope (WCS) to inhibit antibody-antigen interactions, allowing for even diffusion in the tissue. Second, the activity of the WCS is nullified by host-guest reactions with added macrocyclic compounds, restoring antibody activity. New research in Nature Communications introduces a #DeepLearning-based aberration compensation method for #Fluorescence #Microscopy. The authors synthetically introduced aberrations into the raw data and trained the network to reverse them, finding their method similar in effectiveness to conventional #AdaptiveOptics.
