They developed a novel method to assess pericyte shape changes, while showing that MSC-EVs could down-regulate hallmarks of neuroinflammation. As published in Nature Communications, scientists at SHIFTBIO INC. in collaboration with RoosterBio developed SIRP-EVs (engineered mesenchymal stem cell-derived vesicles) as a new treatment for ALF (acute liver failure). This publication highlights the scalable 3D #bioreactor process developed to manufacture high yields of SIRP-EVs coupled with an effective downstream process that concentrates EVs and removes impurities. MSC-derived SIRP-EVs outperform HEK cell-derived versions, highlighting their unique therapeutic potential. In the past decade, extracellular vesicles/exosomes have shown promise for treatment of a variety of diseases and moved into the clinic with dozens of reported clinical trials globally. As the number of clinical trials increases, exosome production requires robust, scalable, and cost-effective processes. 📑 This study led by McGill University's Professor Lisbet Haglund, analyzed #extracellularvesicles (EVs) from intervertebral disc (IVD) cells across tissue degeneration grades. Proteomic analysis revealed 88.6% of EV proteins were shared, but their abundance varied. That is, mildly-degenerate EVs had distinct extracellular matrix-related cargo. Enabled in part by RoosterBio's EV collection medium (RoosterCollect™-EV), these findings shed light on IVD degeneration mechanisms, suggesting biomarkers and therapeutic targets for low back pain.
