#CustomerPublicationWeekly The Sweet Kill? Peripheral blood #immune cells playing a pivotal role in the ongoing surveillance of #cancer cells. Using #singlecell #multi-omics method that can simultaneously detect transcriptome, T cell receptor (TCR) repertoire, and LacNAc glycan profiling, the immune cells in the blood of bladder cancer patient were analyzed. This study revealed significantly higher levels of LacNAc on the peripheral T cells from early-stage bladder cancer patients. Furthermore, T cells with elevated surface LacNAc levels demonstrate enhanced cytotoxic activity and greater TCR clonal expansion. These results highlight LacNAc as a potential surface marker of effector T cells in bladder cancer. Congratulations to to Univ.-Prof. Dr med. Dr h.c. Sergiu Groppa, MBA and team for their recent publication in in Signal Transduction and Targeted Therapy! The study utilized single-cell RNA sequencing (scRNA-seq) on over 101,000 individual cells, uncovering significant transcriptomic changes in glial cell types within the Parkinson's Disease (PD) cortex. By integrating these molecular findings with electrophysiological data, the authors revealed "pan-glial" immunometabolism dysregulation as a central pathological mechanism. These pathways present promising targets for novel immunotherapeutic strategies in PD. Excited to see how this research paves the way for further breakthroughs! #CustomerPublicationWeekly Persistent H. pylori infection triggers the repair program of the mucosa, such as spasmolytic polypeptide-expressing metaplasia (#SPEM). An increase in telomerase reverse transcriptase (TERT) protein expression is observed in chief cells upon infection with cagA-positive H. pylori and TERT knockout significantly ameliorated H. pylori-induced SPEM. #single-cell RNA sequencing revealed suppressed Wnt/β-Catenin pathway in gastric cells with TERT knockout. CagA appears to elevate TERT abundance by disrupting the interaction between TERT and its novel E3 ligase, SYVN1. This study sheds lights on mechanism underlying which CagA activated the TERT/Wnt/β-Catenin pathway and provides molecular basis for targeting CagA-activated Wnt signaling in chief cells for the treatment of gastric precancerous lesions. #CustomerPublicationWeekly Immune infiltration within the tumor microenvironment (#TME) is important for immune checkpoint blockade (#ICB) therapy to function. Mild photothermal therapy (PTT) is often combined with immunotherapy to enhance efficacy, likely by increasing immune cell infiltration and stimulating T-cell activity. Notably, the PTT also accelerates the release of tumor cell-derived exosomes (TEX) and upregulates PD-L1 expression on both tumor cells and TEX. A multi-stage drug delivery system is designed that concurrently delivers photosensitizers (reduced graphene oxide nanosheets, NRGO), anti-PD-L1 antibodies, and exosome inhibitors (sulfisoxazole). This local combination therapy remodels the immunosuppressive TME through exosome inhibition and enhanced immune cell infiltration, while also boosting T-cell activity to trigger systemic antitumor immunity- making it a promising approach to further improve immunotherapy efficacy.
