Dear Friends I am really glad to share that I received a competitive career starter grant from #knightsTemplarEyeFoundation (Marci Martinez) for 1 year of research focused on Leber’s Hereditary Optic Neuropathy (LHON). I would like to thank my mentor Prof. Don Zack for all the guidance and support, all my lab members specially Cindy Berlinicke, Casey Keuthan, Srinivasa Sripathi and my family and friends. Thanks to Prof. Alice Pébay AM for agreeing to share the LHON-iPSCs lines. LHON is the most common form of childhood optic neuropathy; it leads to central vision loss, with the onset ranging from ten to thirty years of age. More than 90% of LHON is linked with mutations in the mitochondrial genes encoding the subunits of NADH dehydrogenase. Despite the ubiquitous expression of these LHON-associated mitochondrial enzymes in all the cells, it lead to preferential dysfunction and death of only Retinal Ganglion Cells (RGCs). With the advancement of stem cells technology patient's specific iPSCs can be differentiated into various cell type including RGCs and other retinal cells. In this project we are going to explore the preferential loss of LHON-RGCs compared to other retinal cells using retinal organoids. We expect the proposed study will help us to understand the potential drivers of LHON-associated RGC dysfunction and cell death; pathways which could then be targeted for the development of LHON therapeutic strategies. https://lnkd.in/e_Yz_xDC
