Hello, I'm Patrick Schramm, Research Scientist and Product Manager at Advanced Targeting Systems. Today we're going to explore more of the fascinating world of pH sensitive fluorescent dyes. These guys are like magical color changing potions that help us see and understand the pH of substances around us. Let's dive right in. PH sensitive fluorescent dyes change color based on the pH level of the solution they're in, as evident in their name. When these dyes are exposed to different pH levels, they undergo a magical transformation. They absorb light of one color and emit light of another, giving us a visual clue about the H At Advanced Targeting Systems, we have a catalog of secondary antibody conjugates with pH sensitive dyes, which we've termed as our fast products. These products can be used with various species, antibodies, or you can even conjugate A biotinylated protein using Streptavidin FAST. The interior environments of cells have a relatively low or acidic pH compared to outside of a cell, specifically the endosomes and lysosomes. When the fast die is attached to another molecule like an antibody, it allows us to track and visualize the journey from the extracellular world into the cells interior as evident in this time lapse video. The fast conjugate takes us on a vivid tour of the macroscopic world of cell biology. It's like a key that unlocks the secrets of cellular compartments and processes. At higher magnification you can notice the small circular dots or punctate structures that are bright. These are the endosomes or lysosomes that currently have the fast molecule inside the cell. In these images and experiments, the only way for the FAST molecule to get inside the cell was via receptor mediated endocytosis. In other words, we attached a primary antibody to a fast conjugate that targeted a cell surface receptor unique for these cells. The positive fluorescence is proof of antibody internalization. This tool is very helpful and effective in understanding how antibodies interact with receptors and cells that you are working on. To sum it up, pH sensitive fluorescent dyes or are fast products gives insight into the invisible world of pH and antibody internalization and it will open doors to various discoveries and innovations.
