Bhawna Dixit's paper is out in Journal of Molecular Biology, a collaboration with Wim Vranken (VUB)! We investigated how well the popular AlphaFold model matches with others models, such as molecular dynamics simulations or structural models derived from experimental data. Specifically, we checked how well the flexibility matches. The answer is: it depends. Outspokenly rigid vs. flexible regions do well. But subtleties in flexibility get lost in the AlphaFold models. My Caltech Center for Science, Society, and Public Policy Co-Director, Frederick Eberhardt, recently talked to John Marken (biology postdoc at Caltech) and Richard Murray (professor at Caltech) about our Center's workshop on the "Pathways Towards the Safe and Effective Deployment of Engineered Microbial Technologies." The workshop was amazing, bringing together people from industry, the policy and regulatory community, and academia. Through collaborative discussions and the sharing of innovative methodologies, these sessions outlined frontier approaches in macromolecular crystallography. This review provides an overview of this rapidly moving field in light of the rich dialogues and forward-thinking proposals explored during the congress workshop and microsymposium. Imagine you have tiny building blocks (proteins) that sometimes stick together in a way that isn't good and can make your brain not work well. In this work, researchers found that when calcium is around, these blocks stick together in a different way, making possible to our brain not to become sick (Alzheimer’s Disease). From MBoC Design principles of Cdr2 node patterns in fission yeast cells Opalko et al. found a new role for the nucleus in positioning cytokinetic ring precursors. They combined particle-based simulations with experiments including anucleate and multinucleated cells. The work has implications for spatial patterning in biology. https://lnkd.in/ghyN85z3

What Defines a Cell Type? The Complexity of Categorizing Cells Tiago Lubiana just earned his PhD! In his thesis, he tackled a question that sounds simple but is actually incredibly complex: What defines a cell type? Estimates for the number of cell types vary widely, from around 200 to over 6,000, depending on the criteria used. Take human T cells, for example. They express CD3 proteins, and if they also express CD4, we call them CD4 T cells. Add a few more markers, like CD25 and FOXP3, and those CD4 T cells become classified as regulatory T cells (Tregs). But now, with single-cell technologies, we can measure hundreds of potential markers, creating even more subcategories. So, where do we draw the line? How do we decide when one cell is distinct enough to be considered a different "type"? Just like defining species, figuring out what makes a "cell type" will eventually need a consensus on clear criteria from the scientific community. And this is especially important in single-cell analysis, which relies on these definitions to map out the diversity of cells in complex tissues and systems.
