Congratulations to the winners of the 2024 Nobel Prize in Physiology or Medicine, Victor Ambros and Gary Ruvkun, for the discovery of microRNA and its role in post-transcriptional gene regulation! While microRNA (miRNA) has been around since 1993, its role in gene regulation still impacts the way scientists conduct their research every day. To briefly explain what microRNAs are, here is a metaphor: Our bodies function like traffic systems, with microRNA acting as traffic lights and gatekeeper genes or transcriptional factors as the police. Just as traffic lights regulate flow automatically, miRNA controls gene expression after protein transcription. If miRNA regulation fails—like a broken stoplight—the police regulators step in, but less effectively. When miRNA malfunctions, often due to issues in protein transcription, it can lead to diseases like heart disease. https://lnkd.in/eFU7DuZf

The 2024 Nobel Prize in Physiology or Medicine has been awarded today to Victor Ambros and Gary Ruvkun for their discovery of microRNA and its role in post-transcriptional gene regulation. They identified microRNA, a novel class of small RNA molecules that play a crucial role in gene expression control. Their seminal discovery was reported in two papers published back-to-back in Cell in 1993 (Lee, Feinbaum and Ambros, 1993; Wightman, Ha and Ruvkun, 1993). It is now known that the human genome encodes over a thousand microRNAs, adding a whole new layer to the complexity of gene regulation. This year’s medicine laureates Victor Ambros and Gary Ruvkun discovered a new principle of gene regulation, mediated by a previously unknown type of RNA, microRNA. They published their results in 1993 but were initially met with almost deafening silence from the scientific community. Although the results were interesting, the unusual mechanism of gene regulation was considered likely irrelevant to humans and other more complex animals. That perception changed in 2000 when Ruvkun's research group published their discovery of another microRNA that was highly conserved and present throughout the animal kingdom. Ruvkun's article sparked great interest, and over the following years, hundreds of different microRNAs were identified. Today, we know that there are more than a thousand genes for different microRNAs in humans, and that gene regulation by microRNA is universal among multicellular organisms. Intriguingly, a single microRNA can regulate the expression of many different genes, and conversely, a single gene can be regulated by multiple microRNAs, thereby coordinating and fine-tuning entire networks of genes.
