Nearly 2 million people are diagnosed with colorectal cancer globally each year, and 20-30% of CRC patients have metastatic disease at the time of diagnosis.    Metastatic / Stage IV colorectal (bowel) cancer occurs when cancer spreads to another part of the body outside the colon, rectum, and nearby lymph nodes. Researchers at Washington University School of Medicine have developed a personalized neoantigen DNA vaccine targeting triple-negative breast cancer. In a phase I trial involving 18 patients, the vaccine was well-tolerated and elicited immune responses in 14 participants. Notably, after three years, 16 patients remained cancer-free. Yesterday, our collaborators from The Institute of Cancer Research and The Royal Marsden NHS Foundation Trust presented groundbreaking data at #ASCO24. In a study of 78 patients with breast cancer (23 TNBC, 35 HER2+, 18 HR+ and 2 unknown), our ultra-sensitive NeXT Personal® assay found that 100% of patients with no detectable residual disease in repeat testing did not have cancer recurrence. ctDNA detection correctly identified disease recurrence with a 15-month median lead time in all 14 patients that relapsed. What are the implications of the main findings❓ 🔹 Clinical approaches to breast cancer treatment could be improved by evaluating somatic mutations in the KEAP1-NRF2 complex in tumourtissue and surrounding benign tissue. This could enhance prognostic accuracy and treatment planning. The variability of genotypes, particularly in histologic grade 2 breast cancer, suggests that understanding genetic differences could be crucial for developing targeted interventions and improving patient outcomes.

A Portrait of Germline Pathogenic Variants in Breast Cancer Genes in Brazil The largest study of its kind in Brazil’s private health system analyzed germline variants in 13 high and moderate penetrance breast cancer genes. Pathogenic variants were found in 9.7% of cases, with 60% in BRCA1/BRCA2. Higher rates were observed in patients under 50 (12%), triple-negative breast cancer cases (14%), and triple-negative patients under 35 (17%). More men die of lung cancer than the next two most deadly types of cancer for men (prostate and colorectal) combined. More women die of lung cancer (an estimated 59,280 deaths in 2024) than breast cancer (an estimated 42,250). Awareness is the first step to prevention.
