🌟 The quality of nucleic acid analysis experiments heavily relies on amplification: 🧬 PCR is commonly used for highly specific amplification of one or multiple targets, with each target confined by a pair of primer hybridization sites. 🧬 For whole genome amplification, isothermal approaches like MDA are generally preferred. Isothermal methods rely on strand-displacing polymerases to enable primer binding and initiate reaction. Partitioning samples into water-in-oil emulsion is a widely recognized strategy to increase signal-to-noise ratio. Research has shown that MDA in droplets tends to outperform bulk reactions, thanks to template molecule isolation, leading to better sensitivity and amplification uniformity. Amplification sensitivity and uniformity are of critical importance for single-cell sequencing. It's crunch time, and our team is fully hands-on as we push to meet our targets. After some delays, we're finally receiving our much-needed lab chemicals, consumables, and equipment, allowing us to ramp up our work. We're running multiple experiments concurrently to answer our own scientific questions, all while preparing for review by our panel. Next up: plasmid extraction, where the purity of plasmids is crucial for successful in vitro transcription and mRNA synthesis. Usually, labs use commercial kits to streamline this process, sometimes we turn to classic methods from the 1960s when resources are tight. Precipitating pDNA in cold alcohol is one of my favorite lab moments – being able to physically see the DNA is a satisfying experience in molecular biology, where so much of our work is invisible and requires verification at every step. After precipitation, it’s onto centrifugation and reconstituting the dry pellets in the buffer, which ultimately results in what looks like just another drop of water. But in that drop lies the foundation of the work we’re doing – a tangible step in the complex journey of molecular research. Exciting times ahead!
