The Packaging and Distribution Expert Committee is proposing the following revisions which will update and expand the scope of the current chapter. Listed below are the key changes being proposed: 1. Change the title to “Elastomeric Components Used in Injectable Pharmaceutical Packaging/Delivery Systems”. 2. Emphasize the baseline requirements for the selection of thermoset and thermoplastic elastomeric components. 3. Expand the scope to include all elastomeric components used in an injection packaging system. Elastomeric components include, but are not limited to, those used for vials, bottles, prefilled syringes (plungers, needle shields, and tip caps), cartridges (plungers and seal liners), injection ports for flexible bags and infusion sets, and plungers for single-use syringes. 4. Delete the Heavy Metals ⟨231⟩ testing and add a modern method for extractable element determination. 5. Omit functionality tests and assessment from the chapter and move them to new chapters appearing in this issue of PF. a. Functionality tests appear in Elastomeric Closure Functionality in Injectable Pharmaceutical Packaging/Delivery Systems ⟨382⟩. b. Baseline information for the assessment is provided in Assessment of Elastomeric Closure Functionality in Injectable Pharmaceutical Packaging/Delivery Systems ⟨1382⟩. 6. Develop a new informational chapter, Elastomeric Evaluation of Elastomeric Components Used in Pharmaceutical Packaging/Delivery Systems ⟨1381⟩, that is meant to support the current chapter revision by: a. Describing elastomeric components and their materials of construction for use in pharmaceutical packaging systems b. Providing a high-level introduction to elastomer chemistry, manufacturing technology, and the post processing of components c. Explaining basic functional characteristics of components d. The Tablet Manufacturing Process. The tablet manufacturing process is a critical step in the pharmaceutical industry. Tablets are a popular form of medication because they are easy to take and easy to store. However, making tablets is not as simple as just putting powder into a mold and pressing it into shape. There are several steps involved in the tablet manufacturing process, and each step must be carefully controlled to ensure that the final product meets all safety and quality standards. manufacturing process The first step in the tablet manufacturing process is pre-formulation. During this phase, the active ingredients and excipients (inactive ingredients) that will be used in the tablet are mixed together in the correct proportions. The mixture is then tested to ensure that it has the proper physical and chemical properties. The next step is the formulation, which is when the actual tablet is created. The mixture from pre-formulation is placed into a tablet press, which forms it into the desired shape and size. Once the tablets are formed, they are usually coated with a film to make them easier to swallow or to prevent them from disintegrating too quickly in the body. After coating, the tablets undergo final testing to ensure that they meet all quality standards. Once they have passed all quality testing, the tablets are packaged and shipped to pharmacies and other customers.

Granulation is a critical process in the pharmaceutical industry, primarily used to improve the properties of powders in the production of tablets and capsules. There are several granulation techniques, each with its advantages and specific applications. The main granulation methods include: 1. **Wet Granulation**: - **High Shear Granulation**: Uses high-shear mixers to blend powders with a granulating fluid, forming dense granules. It's fast and effective for producing uniform granules. - **Fluid Bed Granulation**: Involves suspending the powder in a fluidized bed and spraying it with a granulating solution. It ensures good mixing and uniform granule formation. - **Spray Drying**: Converts a liquid feed into dry granules by spraying it into a hot drying chamber. It’s suitable for heat-sensitive materials. 2. **Dry Granulation**: - **Slugging**: Involves compressing the powder into large tablets (slugs), which are then broken down into granules. It's suitable for moisture-sensitive materials. - **Roller Compaction**: Compacts powder between two rollers to form sheets, which are then milled into granules. It's efficient and used for materials that cannot be processed with wet granulation. 3. **Melt Granulation**: - Uses a meltable binder to agglomerate the powder. The binder is melted, mixed with the powder, and then cooled to form granules. It’s beneficial for improving the dissolution rate of poorly soluble drugs. 4. **Spray Congealing**: - Involves spraying a molten mixture of drug and excipients into a cooling chamber, where it solidifies into granules. This method is suitable for creating sustained-release formulations. 5. **Hot Melt Extrusion**: - Combines powder with a polymer and passes it through an extruder, where heat and pressure create a homogeneous mixture that solidifies into granules. It’s used for modifying drug release profiles. Each of these granulation techniques has specific advantages and is chosen based on the properties of the active pharmaceutical ingredients (APIs) and the desired characteristics of the final product.
