Exactly 10 years ago, we submitted our first paper on ionic liquids to PNAS where we reported the first ionic liquid that we invented, choline-geranate (Cage), for transdermal delivery of antibiotics. Little did we anticipate that it was just the beginning! Over the last ten years, ionic liquids have been demonstrated to deliver many classes of therapeutics including small molecules, peptides, antibodies and nucleic acids by multiple routes including transdermal, oral, and parenteral, among others, for applications in dermatology, oncology, metabolic diseases, infectious diseases, dental hygiene, and many others, with several programs already in clinical trials. Grateful to and proud of the generations of graduate students, postdocs, and academic/industry collaborators/champions who have helped establish ionic liquids as a key enabler of next generation therapeutics. Polychromasia in Hematology: Polychromasia refers to the presence of abnormally high numbers of Reticulocytes (immature red blood cells) in a peripheral blood smear (PBS). Reticulocytes appear larger than mature RBCs, with bluish-gray color when stained with Romanowsky-type stains (e.g., Wright's or Giemsa stains) and leading to a higher MCV. This coloration is due to residual RNA and other organelles in the cytoplasm, characteristic of immature erythrocytes. Reticulocyte Count and Staining: The reticulocyte count measures the percentage of reticulocytes in the blood, which helps assess bone marrow function and response to anemia. For this purpose, 100 microliters of whole blood sample is mixed with 100 microliters of Brilliant Crystal Blue color or New Methylene Blue (equal ratio) and placed in a Bain-Marie or incubate at 37 degrees for 20 minutes. Then a slide is prepared. With a 100 lens , the number of reticulocytes is counted in 10 randomly selected microscopic fields and finally the percentage of reticulocytes is reported. The normal range for the reticulocyte count without anemia is: - Adults/elderly/child: 0.5% to 2.0% - Infant: 0.5-3.1% - Newborns: 2.5% to 6.5% It is better to report the reticulocyte production index (RPI) instead of the reticulocyte percentage as follows: Reticulocyte Index = Reticulocyte count % x Hematocrit / Normal Hematocrit (Normal Hematocrit most commonly accounted as 45%) RPI = Reticulocyte Index/ Maturation correction HCT Retic survival (days) =Maturation correction 36- 45 1 26-35 1.5 16-25 2 <15 2.5 OR RPI = Reticulocyte count x Hemoglobin / Normal Hemoglobin x 0.5 Normal RPI varies between 0.5 and 2.5%. #Hematology #ReticulocyteCount #Polychromasia #AnemiaDiagnosis #RPI #BloodSmear #CBC #BoneMarrowFunction #Erythropoiesis #LaboratoryScience #MedicalLab #LabTesting
