FIG . The indole test. A positive indole test. The 48-hour Escherichia coli culture grown at 37°C tests positive for the presence of indole as indicated by the red reagent layer after the addition of Kovács reagent. The reagent appears as a thin yellow layer on top of the culture medium. See Indole Test Atlas images of indole test using tryptone broth. The last steps to determine the E.Coli is Biochemical test including Methyl Red and (or )Indole test.

Pus sample collected from wound received for CS. Colony Morphology; Dull, non hemolytic, flat growth appeared on blood and Chocolate agar. Biochemical tests; catalase positive, oxidase negative, Non-motile gram positive rods. Gram staining shows spore forming bacteria. Penicillin was resistant to the mentioned spp.
