What are the methods for detecting pyrogens? Several pyrogen detection methods include the RPT (Rabbit Pyrogen Test), MAT (Monocyte Activation Test), and LAL (Limulus Amoebocyte Lysate) assay. The LAL assay has gained widespread acceptance by regulatory bodies such as the FDA and international pharmacopoeias. Among LAL methods, the gel clot method is the most commonly used. This test relies on a coagulation reaction between the endotoxin and the lysate to determine its presence or absence. Did you know deutetrabenazine (Austedo) used for tardive dyskinesia and Huntington disease-associated chorea has PGx guided dosing? From the drug label: "In patients who are poor CYP2D6 metabolizers, the total daily dosage of AUSTEDO should not exceed 36 mg (maximum single dose of 18 mg)." Daily Critical Care Pearls #153: Antimicrobial Adjustments in CRRT 💉The most common of CRRT is continuous veno-venous hemodiafiltration (CVVHDF) in which both diffusion and convection are used. The NDA submission includes data from the Phase 3 APHENITY trial, recently published in The Lancet. The study showed that 60% of patients achieved protein intake above age-adjusted recommendations while maintaining phenylalanine (Phe) levels < 360 μmol/L, demonstrating the potential for improved dietary flexibility.
