I am pleased to share our latest publication on the mechanochemical synthesis of pharmaceutical co-crystals at a kilogram scale. Unlike traditional solution-based methods that typically require large amounts of solvents and energy, our approach uses a drum mill from RETSCH. This demonstrates the potential of repurposing common industrial milling equipment for sustainable large-scale chemical transformations, which aligns with the industry's shift toward greener processes. Key highlights from our findings: -The reaction was completed within 90 min, yielding 99% of pure Ibuprofen:nicotinamide co-crystals without the need for additional purification. -Minimal metal contamination from abrasion was observed, well within acceptable regulatory standards for daily intake. Structurally complex fine chemicals are crucial in the pharmaceutical and agrochemical industries. However, synthesizing them, especially those with chiral centers, poses enormous challenges due to the need for catalyst-supported steps and high product purity. Controlling stereoselectivity during synthesis is vital to avoid costly stereoisomer separation. Current batch processes have long changeover times and complex controls. As a consequence, Fraunhofer IMM and its partner institutes develop a new modular technology platform to address these challenges, focusing on continuous flow synthesis under mild and selective conditions. Initial targets include chiral alcohols with functional groups impacting active pharmaceutical ingredients (API) bioavailability or allowing further selective functionalization.
