Thrilled to present my work at the #RSCPoster2024 LinkedIn Conference-2024 on the topic “Organocatalyzed diastereoselective cyclization of β-alkyl nitroolefins with alkylidene malononitriles: new approach to azetidine nitrones and isoxazoles.” Here, we have developed the methodology to synthesize highly strained azetidine nitrones with a tetra-substituted chiral carbon centre as well as fully substituted isoxazoles. We have treated β-alkyl nitroolefins and β-aryl/heteroaryl/alkyl-substituted alkylidene malononitriles in presence of a catalytic amount of DABCO at room temperature. After 1-2hour diastereoselective azetidine nitrons have formed with good to high yield and excellent group tolerance. We have successfully synthesized 33 substrates with a good diastereomeric ratio (up to ≤ 93:7). Further, by using inexpensive Brønsted acid catalyst in this reaction for 12 hours, a novel class of fully substituted isoxazoles have been formed with 24 examples. "I have been using #Cryolocks for years and recently published them in a paper on my 'Fast & Furious' vitrification method. This study showed that immature oocytes vitrified and warmed using a short protocol survived and resumed meiosis without compromising survival rates. Additionally, it significantly reduced the time needed for vitrification and warming. Cryolocks are easy to handle and load, reliable, robust, and easy to label using a Brady labeler.
