Molecule of the year -3 (2024) One hundred years ago, Julius Bredt's observation that certain molecules couldn't form double bonds between adjacent carbon centers, known as "anti-Bredt" olefins, but now has been challenged by McDermott et al., who report a strategy to prepare these olefins as fleeting intermediates for cycloaddition reactions. Rule-bending strained alkene A solution to the anti-Bredt olefin synthesis problem Bridged organic molecules with a double bond at the bridgehead position (shown here as two lines in red) have been regarded for 100 years as too strained to make, but that hasn't stopped chemists from trying. This year, they finally succeeded (Science 2024, DOI: 10.1126/science.adq3519). Breaking a Century-Old Rule: Chemists Achieve the 'Impossible' with Anti-Bredt Olefins (ABOs)! 🌟 As a chemist, I'm always fascinated by the mechanisms behind molecules, diving into the details, and following the references (thanks to Dr. Radhakrishnan KV, Dr. Anilkumar Gopinathan and Dr.Preethanuj Preethalayam for introducing me such fascinating bridged structures). So, when I saw this latest breakthrough (https://lnkd.in/gJAxDp7y), I was blown away! Publishing research like this is no easy feat, yet these scientists have done the "impossible." For the first time, a team has managed to create anti-Bredt olefins (ABOs)—a type of molecule scientists thought too unstable to exist. ; With these notoriously tricky molecules, researchers now have a brand-new path to crafting complex compounds. This could even help in creating drugs that were previously out of reach! Craig Williams from the University of Queensland calls it a “landmark contribution,” and I completely agree. This achievement, published in Science, is a huge win for innovation and curiosity-driven science. Straightforward Path to Cyanoformamides Author: ChemistryViews courtesy: Chemistry Views Cyanoformamides (RHN–C(=O)–CN) are useful intermediates in organic synthesis, e.g., for the preparation of heterocycles. However, their synthesis can require toxic CN-containing species and other hazardous reactants. Vittorio Pace, University of Vienna, Austria, and University of Turin, Italy, and colleagues have developed a method for the straightforward preparation of cyanoformamides that is based on cyanide release from a suitable precursor in the presence of electrophilic isocyanates. The team used PhMe2SiCN as a cyanide “reservoir”, which releases CN– at a fast rate upon the formation and collapse of a complex with the Lewis base potassium tert-amylate (KOtAm). The researchers reacted a variety of widely available isocyanates with different aromatic or aliphatic substituents with PhMe2SiCN in the presence of catalytic amounts of KOtAm in tetrahydronfuran (THF). The desired cyanoformamides were obtained within 1 min in high to excellent yields. Violaceoids A-F and eutyscoparols A-G are a group of bioactive compounds recently isolated from fungi that are believed to have promising antimalarial and antibacterial properties. Since their discovery, there have been efforts to synthesize them in the laboratory in sufficient quantities for further study. In a study featured on the cover of Volume 13, Issue 7 of the Asian Journal of Organic Chemistry, scientists from Tokyo University of Science (TUS) have successfully synthesized these compounds from dinitriles on a subgram scale. The proposed approach takes about nine steps and increases the yield to about 30%. The method is an improvement over previous methods and can lead to the development of new drugs. "Catalyst-Free, Three-Component Synthesis of Amidinomaleimides" 📚 Summary 👨🏫 A recent study from The Journal of Organic Chemistry, from American Chemical Society - Division of Organic Chemistry presents a new catalyst-free three-component reaction for synthesizing amidinomaleimides. Thanks to the authors from the Gregory Weiss team from UC Irvine. Mechanism Insights 🔬 The reaction proceeds through a 1,3-dipolar cycloaddition, starting with the formation of an enamine from the secondary amine and aldehyde. This enamine then reacts with the electrophilic azidomaleimide, forming a triazoline intermediate, which subsequently undergoes ring-opening to yield the amidinomaleimide product.
