Organic chemists always want more options for making carbon-carbon bonds. Experience has taught many of us that best laid plans can go to waste – “hope for the best but prepare for the worst” is my mantra. Today, we also understand the importance of environmental protection and sustainability, we want alternative methods that are green and one place to start is developing processes that don’t use heavy rare earth metals such as palladium. You can be sure that the scarcer a metal is, the more energy demanding it’s extraction. I am therefore always happy to see new synthetic methods that traditionally used palladium, use something else – such as nickel. So I was excited to see this nickel-catalysed carbonylative Negishi cross coupling published just last week: https://lnkd.in/e2_NXbfz. The reaction stitches together a secondary alkyl group (alkyl iodide precursor) to carbon monoxide and a primary alkyl group (organozinc precursor). The reaction conditions are straightforward and rely on the use of common NiCl2.DME, ligand and lithium chloride/iodide in dimethylacetamide. The ligand requires a one step synthesis – amide coupling using iso-butylchloroformate and for purification, an alumina column with a petroleum ether – dichloromethane – ethyl acetate solvent system. Maybe a bit of room for improvement here. The reaction works in the presence of esters, protected alcohols and amines, alkenes, acetals and even a pinacol borane and does offer a genuine alternative to the traditional Grignard addition to Weinreb amide or much more modern reaction between ester and sodium alkylsulfinate: https://lnkd.in/enwR_JUT - although this one will need some further development to reach the heights of the Negishi technology. As someone who is interested in what reactions can do rather than developing the methodology, I can sometimes find the mechanistic discussions dry and a little bewildering. Here, I thought the authors did a great job explaining their radical trapping/clock and stoichiometric experiments and translating the results into a plausible mechanism that I could understand.
