Established diagnostic ions for structural elucidation and proposed fragmentation pathways, including competition between CH<sub>3</sub>· and methane losses in rhamnetin and isorhamnetin.

Breath Meets Silver for Better Untargeted Metabolomics! 🌬️🔬 A study conducted by Cedric Wüthrich, Renato Zenobi, and Stamatis Giannoukos from ETH Zurich considered Silver Nitrate (AgNO3) as an alternative dopant for untargeted breath metabolomics using (SESI-Orbitrap). This approach has revealed new aspects of detecting and analyzing biomarkers in exhaled breath. Results: ● Enhanced Ionization and Detection: AgNO3 improves ionization for compounds such as unsaturated hydrocarbons and sulfur-containing compounds, and reveals unique elemental ratios, particularly for low oxygen-carbon and high hydrogen-carbon ratios. ● Comprehensive Feature Coverage: AgNO3 provided the most comprehensive coverage of compounds in exhaled breath, identifying 1679 unique features. In comparison, aqueous FA detected 708 unique features, and NaI detected 531 unique features. ● High Molecular Mass Detection: Features detected using silver-doped electrospray possessed higher molecular masses and were more numerous compared to other conditions, leading to distinct separation in principal component analysis (PCA) plots. Targeted protein degradation Targeted protein degradation involves redirecting protein recycling systems in our cells to destroy the disease-causing proteins. Protein degraders work by capturing the disease-causing protein and making it stick like a glue to the cellular protein-recycling machinery, which then tags the protein as expired in order to destroy it. The tag is a small protein called ubiquitin, which effectively gets fired at the disease-causing protein like a bullet. In order for the process to work effectively, ubiquitin must hit the right spots on the target protein so that it gets tagged effectively. The new work by the team enables them to see how the bullet hits the proverbial bull’s eye. Working with a protein degrader molecule called MZ1, and using high-end mass spectrometry, they were able to identify exactly where on the target protein the vital ‘tags’ are added. The work shows how degrader drugs hold onto and position disease-causing proteins, making them good targets for receiving ubiquitin molecules (ie. The health benefits of Baobab powder keep getting more exciting every day! A recent study using advanced Metabolomics identified 40 metabolites for the first time. The study also revealed the antidiabetic potential of Baobab with this abundance of bioactive phytochemicals. Histopathological and immunohistochemical studies demonstrated that Baobab exerted hepatoprotective and renoprotective effects in diabetic rats. Lately, I've been diving deep into the world of enzymes used in the clinical diagnostic laboratory, particularly proteinase K. This simple broad-spectrum serine protease acts as a tiny scissor-like enzyme, efficiently cutting proteins into smaller pieces by snipping the connections between their peptide bonds. In the molecular labs, it plays a crucial role in eliminating unwanted proteins and inactivating enzymes like RNase during nucleic acid extractions needed for downstream assays.
