On September 6, 1978, the production of biosynthetic human insulin using recombinant DNA technology was announced. Scientists at City of Hope National Medical Center and Genentech developed a method for producing human insulin using E. coli bacteria expressing the genes for the two polypeptide chains that make up the hormone. Eli Lilly licensed the method to commercialize it. It took the FDA only 5 months to approve "Humulin" and on October 28, 1982, the first approved medical product produced using recombinant DNA technology was on the market. Palm tocotrienol (also known as tocotrienol-rich fraction, TRF) is touted as Mother nature’s gift to the brain. Why it is being said so? To date, solid research evidence (from in-vitro up to human clinical studies) backed the neuroprotection benefits exerted by bioenhanced palm mixed-tocotrienol complex. How do genetic variations impact the actions of medications? Part I. Pharmacogenes code for medication transporters. Medication transporters are responsible for moving drugs in and out of target tissues, regulating drug exposure to metabolic enzymes and eventually to elimination pathways. An example of a medication transporter is SLCO1B1 which is predominantly located in liver cells. Lipid lowering statin medications rely on SLCO1B1 for uptake into liver cells - where they exert their effects to lower total and LDL cholesterol - and subsequently undergo metabolism and elimination. SLCO1B1 genetic variations are associated with reduced statin uptake by liver cells and delayed metabolism and elimination. Patients with decreased or minimal to no transporter function likely experience reduced efficacy and are at greater risk for developing statin-associated musculoskeletal symptoms (SAMS), as well as more serious adverse effects such as myopathy and rhabdomyolysis. Preemptive pharmacogenetic (PGx) testing, via a simple cheek swab, can determine if genetic variations of transporters such as SLCO1B1 are present. Aardvark Therapeutics is planning to raise up to $200mn in an initial public offering as early as this summer, propelled by investor hype over anti-obesity medications. Aardvark’s lead drug — ARD-101 — has succeeded in suppressing hunger cravings in patients with Prader-Willi syndrome, a rare genetic form of obesity, in early-stage trials. Investors are excited by ARD-101, a so-called TAS2R agonist, because it does not just activate GLP-1, the gut hormone used in popular weight-loss drugs from Novo Nordisk and Eli Lilly and Company, but it also harnesses CCK. This second hormone could limit widely reported side effects of nausea and wastage of lean muscle mass associated with GLP-1 drugs.
