Inflammatory diseases, including #infectiousdiseases, #diabetes -related diseases, #arthritis -related diseases, #neurologicaldiseases, digestive diseases, and #cancer #tumor, continue to threaten human health and impose a significant financial burden despite advancements in clinical treatment. Pyroptosis, a #proinflammatory programmed cell death pathway, plays an important role in the regulation of #inflammation. Moderate pyroptosis contributes to the activation of native #immunity, whereas excessive pyroptosis is associated with the occurrence and progression of inflammation. Pyroptosis is complicated and tightly controlled by various factors. Accumulating evidence has confirmed that #epigenetic modifications and #posttranslationalmodifications (PTMs) play vital roles in the regulation of pyroptosis. Epigenetic modifications, which include DNA methylation and histone modifications (such as methylation and acetylation), and post-translational modifications (such as ubiquitination, phosphorylation, and acetylation) precisely manipulate #geneexpression and protein functions at the transcriptional and post-translational levels, respectively. In this review, we summarize the major pathways of pyroptosis and focus on the regulatory roles and mechanisms of epigenetic and post-translational modifications of pyroptotic components. We also illustrate these within pyroptosis-associated inflammatory diseases. A “gut-first” hypothesis could shape future Parkinson’s Disease treatments, according to a new study published this month in JAMA. Harvard researchers found a connection between H. pylori infection and a higher risk of Parkinson’s disease in patients. The researchers suggest that the bacteria may trigger inflammation in the body, which could contribute to neurodegeneration. This leads to the hypothesis that, for some people, Parkinson’s might start in the gut and spread to the brain through the vagus nerve. Research into the gut-brain axis is crucial considering that Parkinson’s affects over 90,000 people every year, and emphasize the importance of advocating for further research into gut biomarkers and early intervention strategies. It’s encouraging to see research in this area that will hopefully lead to more positive outcomes for those affected by this disease. Read our new blog post, "Dear Biobanks, please update yourself on endometriosis"! Veronika Denner explains how in most studies and biobanks endometriosis is still categorized as a “non-inflammatory disease of the female genital tract", even though we now know that it is a systemic inflammatory disease characterized by the growth of lesions similar but not identical to the native endometrium. In 2022, the WHO finally recognized that this term is incorrect and the ICD-11 classifies endometriosis as “disease of the female genital system.“ This is a step towards the right direction, however, it is still slightly misleading, as 1) endometriosis can grow far beyond the pelvis, and 2) the term "endometrial" is still used instead of "endometrial-like". However, it finally acknowledges endometriosis as an inflammatory disease, includes thoracic endometriosis and more precise codes for bowel endometriosis depending on the exact site of the lesions, and recognizes adenomyosis with a separate ICD code. Now it is essential that biobanks, like the UK Biobank uses the correct term for endometriosis, which will facilitate research and avoid misunderstandings about the disease.
