The #KI (Potassium Iodide) #Discus Test is a method used to test the performance of Class II Biological Safety Cabinets (BSCs) to ensure they are providing proper protection. This test evaluates the cabinet’s ability to contain and remove contaminants, simulating aerosol dispersion. Here’s how it works: Purpose of the KI Test • To verify the containment capabilities of the BSC. • To ensure the safety of personnel, products, and the environment from potentially hazardous aerosols. Procedure Overview 1. Setup: A potassium iodide (KI) aerosol is generated inside the biological safety cabinet using a nebulizer or discus generator. 2. Testing Points: The KI aerosol is released in specific locations inside the cabinet (usually around the working area). A series of samplers are placed: • At the operator’s position. • Around the cabinet’s perimeter. • In other critical areas to detect potential leaks. 3. Sampling: • The samplers contain test filters that attracts KI particles. • If the cabinet is functioning correctly, little to no KI particles should escape to the samplers. 4. Detection: • Palladium chloride is used to facilitate the chemical reaction that detects potassium iodide (KI) particles on the filters, forming dark spots. 5. Evaluation: The dark spots on the filter indicate the presence of potassium iodide particles, signifying a leak from the safety cabinet. Significance • A cabinet that passes the KI test demonstrates its ability to contain aerosols effectively, ensuring operator and environmental safety. • Cabinets failing the test may need adjustments in airflow, filter replacements, or maintenance.
