❣️ Exciting Research in genetics Exploring Protein Extraction! ❣️ Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) is a widely used technique in proteomics and molecular biology for separating proteins based on their molecular weight. Here's an explanation: Principle of SDS-PAGE 1. Denaturation of Proteins: Proteins are treated with SDS, an anionic detergent, which binds to proteins, denaturing them and imparting a uniform negative charge. This ensures that the migration of proteins in the gel is based solely on size, not charge or shape. Proteins are often also heated in the presence of a reducing agent (like β-mercaptoethanol or DTT) to break disulfide bonds. 2. Polyacrylamide Gel: The gel is made of a matrix of acrylamide and bisacrylamide. The pore size in the gel depends on the concentration of acrylamide; smaller pores allow better resolution of smaller proteins. Proteins move through this gel matrix under an electric field. 3. Electrophoresis: When an electric current is applied, the negatively charged proteins migrate towards the positively charged electrode (anode). Smaller proteins move more easily through the gel's matrix, whereas larger proteins encounter more resistance and migrate more slowly. 4. Separation by Size: The migration distance is inversely proportional to the logarithm of the molecular weight. Thus, smaller proteins move farther down the gel. As a final-year botany student, I’m thrilled to be diving into my research on protein extraction from different rose species. Roses aren't just beautiful—they also hold incredible potential in biotechnology and agriculture. My project aims to uncover the proteins that play a role in their growth, resilience, and even their therapeutic properties. By extracting and analyzing these proteins, I hope to contribute to the growing field of plant-based biochemistry and potentially find new avenues for improving plant health, sustainability, and even medicinal applications.
