Last week, I presented at the journal club of the ASA Covariate Adjustment Working Group on the topic of variance estimation for covariate-adjusted estimators of average treatment effects in clinical trials with binary endpoints. A recording of the session is available at https://lnkd.in/eg6ukft5 It's an important topic because following the ICH E9 R1 Addendum on Estimands and last year's FDA Guideline for Industry on Covariate Adjustment, clinical researchers have been given very clear guidance for point estimation of average treatment effects but much less direction on how to estimate the variance of such estimators, which is of course needed to produce test statistics and confidence intervals. The presentation (joint work with Mark Baillie, Alexander Przybylski and Craig Wang) aimed to bring greater clarity on the regulatory acceptability of several proposed variance estimators. We received generous and insightful feedback from invited discussants Ting Ye, Daniel Rubin (FDA) and Dong Xi. Many thanks to Bingkai Wang for organizing the session.
