As a former #educator, I really love when undergraduate organic chemistry comes to life in such an elegant and clever way. This synthesis of (+)-cylindricine B from the Florida State lab of Joel Smith utilizes the following steps: 1. Acylation 2. SN2 dealkylation and subsequent pyridine dearomatization (rationalized via resonance) - particularly clever! 3. Conjugate addition x2 4. SN2 5. Enolate alkylation 6. Thio-SN2 on methyl carbamate and concomitant decarboxylation The second conjugate addition is diastereoselective due to 1,3-diaxial strain in the undesired product transition state. In the presence of TMSOTf, the authors propose a twist-boat confirmation that exacerbates this strain. This diasteroselectivity then confers preference to the remaining reactions to complete the racemic synthesis of a single diastereomer. On this day in 1882, Friedrich Wöhler died. Wöhler was the first to successfully synthesize an organic compound from inorganic reagents. He did so by heating ammonium cyanate and thus create its organic isomer, urea. Wöhler thereby showed that organic substances can be created in a laboratory, which was thought to be impossible previously.
