Day of Immunology 2024 theme is “Immunity Through the Ages: Navigating the Science of Aging and Immunology“. The innate and adaptive #immune dysfunction that accompanies #aging is known as #immunosenescence. This concept was first described in 1964 by Roy Walford as “reduced effectiveness of the immune system”. Vagelis Rinotas and Marietta Armaka at BSRC Alexander Fleming in Greece studied a protein called TNF, which is essential in both normal body functions and diseases like Rheumatoid Arthritis (RA). In RA, too much TNF makes specific cells in the joints, called synovial fibroblasts (SFs), overly active, leading to inflammation and joint damage. The researchers focused on another protein called Cyld, which helps control how SFs respond to TNF. They found that when Cyld doesn't work properly in these cells, it worsens RA by increasing the levels of certain harmful chemicals and enzymes. This happens because Cyld typically helps to limit the overactivity of SFs by controlling specific signaling pathways in the cells. In summary, Cyld is important for keeping SFs in check and preventing the worsening of arthritis caused by too much TNF. Glad to share our recently published study in Inflammatory Bowel Diseases: https://lnkd.in/eFMAtyAC In this study, we applied high-dimensional analyses to profile the immune compartment in CD-associated and cryptoglandular perianal fistulas. Our analyses revealed that activated HLA-DR+CD38+ effector CD4+ T cells with a Th1/17 phenotype were significantly enriched in CD-associated perianal fistulas compared with cryptoglandular fistulas. These cells, displaying features of proliferation, regulation, and differentiation were also present in blood, and colocalized with other CD4+ T cells, CCR6+ B cells, and macrophages in fistula tracts. Targeting HLA-DR and CD38-expressing CD4+ T cells may offer a potential new therapeutic strategy for CD-associated fistulas. Many thanks to all co-authors for their contributions to this study! ✌🏾Our new paper reveals how #microbiome and #immune cues impact on Ventilator-Associated Pneumonia (VAP) in COVID-19. Our article delves into the complex interplay of lung microbiome and host immunity in the development of VAP among critically ill COVID-19 patients. Our data revealed: Microbial Shifts- We identified distinct alterations in the respiratory microbiome, particularly an increase in Staphylococcus and Enterobacteriaceae, in patients who developed VAP. Immune Dysregulation- VAP was associated with higher SARS-CoV-2 viral loads, elevated neutralizing antibodies, and a paradoxical reduction in key inflammatory cytokines (IFN-δ, IL-1β, IL-12p70, IL-18, IL-6, TNF-α, and CCL4). Metabolic Perturbations- Distinct metabolomic profiles were observed in VAP and non-VAP patients, suggesting a potential role for metabolic dysregulation in VAP pathogenesis. These findings have significant implications for understanding, predicting, and potentially preventing VAP in the context of COVID-19. Thanks to our J.
