R&D and Manufacturing of GGFG linker and exatecan faced two challenging issues in the ADC drug production, which should produce irreversible effects on the preclinical study and clinical study. First issue is racemization of the F amino acid during the peptide coupling significantly impacting on the release efficiency of payload. Another problem is the dr value of Exatecan coming from the chemical suppliers or CROs, which shall reduce the efficacy of ADC drug in the clinical study. Very nice work by Carmine D'Amico and colleagues, now out on ChemRxiv as a preprint! We are introducing poly(2-oxazoline) based microneedles for transdermal drug delivery. The patch itself is an amorphous solid dispersion with mechanical properties suitable for effective skin penetration. The amphiphilic triblock copolymer used dissolves directly into drug loaded micelles with high drug loading, we tested here up to 40 wt.%. The drug loading also effects the dissolution rate, potentially enabling slow and controlled drug release. Collaboration with Hélder A. Santos Anna-Lena Ziegler, Marianna Kemell and Giuseppina Molinaro at University of Helsinki and UMCG research.
