Megan Perry joined us 2 months ago. She is a Biochemistry Masters student excited to work within the Douglas group for the next 9 months. Her project is a preclinical evaluation of candidate Epstein-Barr Virus (EBV) vaccine antigens. It has the potential to significantly impact the fight against EBV-related diseases. Megan is passionate about vaccine development and hopes to continue working in the field after completing her project.

Very happy to have the first half of my PhD project submitted. In the initial part of my PhD, I first established a colorimetric assay in the presence of lipids, compatible with high throughput screening (#HTS) to measure enzymatic activity for EHD proteins. In this way, we identified a drug-like molecule that inhibited EHD4’s liposome-stimulated ATPase activity. Our structure-activity relationship (SAR) studies indicate sites of preferred substitutions for more potent inhibitor synthesis. Thanks to the screening unit at FMP where I did my drug screening and all the co-authors for their contribution. Thanks to the online scientific community for a lot of suggestions and discussions. Pyrazole derivatives have shown promising therapeutic potential due to their diverse biological activities. The synthesis involves the preparation of various pyrazole derivative using multi-step organic reactions, followed by thorough characterization using spectroscopic techniques such as NMR, IR spectroscopy. The anti-cancer and anti-inflammatory activities of the synthesized compounds will be evaluated through in vitro and in vivo assays.The results obtained from this study aim to contribute to the development of new therapeutic agents for cancer and inflammatory diseases.
A team of researchers at Brown University has developed a way of using machine learning to rapidly predict multiple protein configurations to advance understanding of protein dynamics and functions. Our study exploring protein alternative conformations 𝐚𝐧𝐝 their relative populations with modified AlphaFold2 is up in 𝘕𝘢𝘵𝘶𝘳𝘦 𝘊𝘰𝘮𝘮𝘶𝘯𝘪𝘤𝘢𝘵𝘪𝘰𝘯𝘴! In this work, we show that AlphaFold2 can be adapted to predict protein ensembles whose relative state populations strongly correlate to experiment. This approach has significant impacts for drug discovery and biophysics by drastically accelerating the rate at which protein conformations can be explored—from months to mere hours! In our flagship example, we show this to be the case for the Abl1 kinase and its orthologs, as well as its activating and inactivating mutants, with remarkable accuracy and speed. Beyond Abl1, we benchmarked our approach by applying it to nine other protein systems with interesting and contrasting dynamics. Check the supplementary information for a full list! We believe this approach could be invaluable for predicting relative state populations at scale, discovering new druggable conformations, probing the effects of mutations, understanding the evolution of pathogens, and more. In a recent review article published by the Rullo Lab, we had the pleasure of discussing some of the innovative approaches we have developed on "Leveraging Covalency to Stabilize Ternary Complex Formation for Cell-Cell Induced Proximity". Our work focuses on enhancing the ability of immune cells to recognize and kill tumour cells more effectively. We achieve this through covalent bifunctionals that improve immune complex stability and target residence time - a strategy validated under different approaches. As the second author, I was delighted to contribute to this review, and I am thrilled to see our work gaining recognition in the renowned Science blog "In the Pipeline" by Derek Lowe. As he noted in the blog : "We are going to learn a great deal from trying this (some of it the hard way, inevitably), but the possible rewards are tremendous". Excited to share my postdoc work now on bioRxiv! https://lnkd.in/gT6S6jRb We discovered a non-genetic drug-adapted state that emerges frequently in the the human fungal pathogen Candida albicans. It is heritable and reversible, with features of protein- and chromatin-based epigenetic inheritance. This previously uncharacterized behavior, which we termed ‘para-resistance’, is induced by exposure to low, subtherapeutic doses of fluconazole, and facilitates the rapid evolution of resistance to high doses of azoles. It also arises spontaneously during passage through the mammalian gut and compromises the in vivo efficacy of therapeutic doses of fluconazole. Our work sheds light on how organisms rapidly acquire drug resistance, with significant implications for predicting and improving therapeutic outcomes in the treatment of infectious diseases, cancer, and other human pathologies. Senior BioAscent scientists, Duncan McArthur and Angelo Pugliese, named as authors on recent paper: ‘Reversible small molecule pan-Ras inhibitors display tunable affinity for the active and inactive forms of Ras’. During their time at the CRUK Beatson Institute Drug Discovery Unit (now Cancer Research Horizons Scotland Institute), Duncan and Angelo worked on a drug discovery project, together with colleagues from Novartis, which led to the discovery of a series of small molecules that, depending on chemotype, preferentially bind and inhibit mutated Ras in either its active or inactive state or both with similar affinity. This study provides exciting new starting points to explore both pan-Ras and GTP-bound active state inhibition. A great deal of teamwork and creative and skilled medicinal and computational chemistry effort was needed to progress from the initial fragment hits. Before taking up new roles at BioAscent, Duncan was the medicinal chemistry project leader and Angelo was the team lead for the computational work.
