Microglia, the brain's resident immune cells, play a crucial role in maintaining brain homeostasis and responding to injury or disease. Our international neurophysiology research consortium has published a summary of the most recent evidence suggesting a contribution of parvalbumin-positive (PV+) interneuron dysfunction to the pathophysiology of a spectrum of brain proteinopathies and primary neurodegenerative diseases, such as Alzheimer's disease, Frontotemporal Dementia (FTD), and Dementia with Lewy bodies (DLB). Specifically, we focused on the interplay between misfolded protein accumulation and this specific subtype of inhibitory interneurons whose functional alteration leads to network hypersynchrony, cortical hyperexcitability, and gamma rhythm disruption, with the aim of integrating these findings into a coherent pathophysiological framework for each disease. Highlights: • Parvalbumin-positive interneurons regulate cortical excitation/inhibition balance. • Their dysfunction results in cortical hyperexcitability and gamma rhythm disruption. • PV+ interneurons are impaired in AD; their restoration improved patients’ cognition. • Albeit with some differences, PV+ cells are also impaired in FTD and DLB mouse models. • Their dysfunction represents a common pathophysiological aspect across etiologies. 📢 #Publication alert 🩸Vascular #dementia (VaD), the second most common form of #dementia (after #Alzheimers disease), is associated with cerebrovascular dysfunction, often as a consequence of chronic vascular damage and reduced blood flow in the #brain. 🐁Dr Latha Diwakar's lab developed a mouse model of VaD. This was done using the vasoconstricting peptide Endothelin-1 (ET-1) to mimic recurrent transient ischemic attacks. The findings show that ET-1 disrupts the blood-brain barrier, leading to neuronal loss and memory impairment.
