Phosphoramidites are key building blocks in oligonucleotide synthesis, allowing for the sequential addition of nucleotides to create synthetic DNA or RNA strands. It is nice articale for methylation of pyrazoles at N1 by using bulky silane methyl chloride!
Hands up if you have ever attempted to selectively methylate the N1-position of a substituted pyrazole with methyl iodide. Let me guess, you ended up with a near equimolar mixture of N1 and N2-substituted products? Yeah, me too… It’s therefore no surprise that research in trying to improve this (lack of) selectivity is an active area. Chemists at Genentech have made what looks to be an important contribution to the field: https://lnkd.in/eKVqZT7F. They have shown that doing the ‘methylation’ with (chloromethyl)triisopropoxysilane (CAS: 18162-82-8) which is commercially available – it is – I checked, facilitates greater selectivity for the pyrazole N1 position due to sterics. Protodesilylation can then be achieved in the same pot – just add water and TBAF to furnish the desired methylated species. You will still need to purify your reaction mixture by column chromatography to remove the unwanted N2 isomer but you’ll hopefully achieve a greater yield that if you used good old-fashioned methyl iodide.
