BD Rhapsody™ System Publication Highlight
Liu N, Zhang J, Yan M, et al. Supplementation with α-ketoglutarate improved the efficacy of anti-PD1 melanoma treatment through epigenetic modulation of PD-L1. Cell Death Dis. 2023; 14:170
This study aims at understanding α-KG’s function in anti-PD1 treatment by using the BD Rhapsody™ Whole Transcriptome Analysis (WTA) Assay alongside BD® AbSeq Antibody Conjugates and BD® Single-Cell Multiplexing Kit.
⏩ What they discovered: The study found that α-ketoglutarate (α-KG) supplementation enhances the efficacy of anti-PD1 melanoma treatment by increasing PD-L1 expression through the activation of STAT1/3 and TET2/3 enzymes, leading to higher levels of 5-hydroxymethylcytosine in the PD-L1 promoter.
⏩ What this means: This discovery suggests that α-KG supplementation could be a promising strategy to improve the effectiveness of anti-PD1 immunotherapy in melanoma, providing new insights into the metabolic regulation of immune responses in cancer.
