Abstract:
Clinacanthus nutans is an herbaceous medicinal plant commonly found in Asia.
This plant is known to have many pharmaceutical and nutraceutical properties and is traditionally used to treat snakebite, herpes simplex virus and cancer.
Despite being commonly used, reports on its agronomic and genetic improvements are scarce.
No published work on the tissue culture of this species is found so far.
Establishment of tissue culture protocol of this species is essential as a platform for crop improvement using biotechnology tools.
In this study, the effects of two different disinfecting agents (Clorox, H2O2) were investigated at different concentration and exposure time.
Leaves undergone pretreatment of 1% carbendazim a week before harvest and sterilization with 30% of Clorox for 30 min gave survival of 96.7%. H2O2 was equally effective but at longer exposure, which is rather time-consuming.
The callus induction for C. nutans was also investigated using NAA and BAP at various combinations.
Result shows that high amount of hormones (16 mg L-1 of NAA and BAP) is required for the induction of callus. C. nutans is prone to contamination.
This research found Colletotrichum sp. fungus endogeneously in the leaves of C. nutans.
